Acute promyelocyte leukemia (APL) is a kind of malignant clone disease in haemopoetic stem cells.Plenty of abnormal archaeoeytes and juvenile cells proliferate in marrow, and infiltrate generally in all kinds of organs and suppress the haematopoiesis.Triptolide, isolated from the herb Tripterygium wilfordii, as well as some of its derivants has been shown to potently induce apoptosis and autophagy in various malignant cells by inhibiting RNA synthesis and nuclear factor-κB activity. In this project, we aim to confirm whether APL cells can be induced to apoptosis and autophagy by triptolide effectively. Furthermore, we also focus on exploring the molecular mechanism of triptolide induced apoptosis and autophagy. The answers to these questions will lay a foundation for the development of new anti-APL drugs.
本项目以急性早幼粒细胞白血病(Acute promyelocyte leukemia,APL)为研究对象,从小分子药物诱导细胞凋亡及自噬的角度入手,以雷公藤甲素衍生物作为小分子干预手段,结合APL发病及耐药的生理病理微环境理论,拟证实雷公藤甲素可有效诱导AML细胞发生凋亡及自噬,评估雷公藤甲素作为APL治疗药物的可能性,为新型抗AML药物的研发奠定基础。
