The research task in this project has been completed and the satisfactory results from the research project were obtained: it is confirmed that genus-specific protein antigens existed on the surface of leptospiral cells, the genes responsible for genus-specific protein antigens were cloned and screened and 3 genotypes of OmpL1 gene and 2 genotypes of LipL32 gene in the 15 strains belonging 15 serogroups of leptospira Interrogans in China were firstly found, the expression systems of genus-specific protein OmpL1 and LipL32 genotypes were constructed, the expression systems of CTB and LTB which used as adjuvants in genetic engineering vaccine were constructed, the products of the OmpL1 and LipL32 genes were able to express in 15 strains belonging 15 serogroups of leptospira Interrogans in China by using microscope agglutination test, the immune protective rates of rOmpL1 and rLipL32 mixed antigens at different dosages in guinea-pigs infection model could reach 83.8%~100% which set up a stable basis for developing genus-specific genetic engineering vaccine of leptospira Interrogans.
提纯钩端螺旋体属特异性抗体并获取抗血清。建立cDNA文库,筛选属特异性抗原基因克隆及构建其表达系统。应用显凝试验和动物实验考核各属特异性融合蛋白的交叉凝集及交叉免疫保护作用。研究结果为研制钩端螺旋体属特异性基因工程疫苗提供科学依据并奠定较好的研究基础,具有较大的学术价值和应用前景。
