Swimming crab, Portunus trituberculatus, is one of the most important marine economic crustacean in China. Ovarian development is essential to keep its high efficient reproduction, but the molecular mechanism for ovarian development regulation is still unknown. In the previous study, the authors compared the difference between matured and unmatured ovary by using method of mating controlling, in which, the authors found that Wnt pathway might be involved in ovarian development. The purpose of the project are to clone the key genes of Wnt signal pathway, such as Wntless, GSK3 and Frizzled. Then the expression of the key genes of Wnt signal pathway at different stage of ovarian development are to be studied. In addition, role of Wnt signal pathway on ovarian development will be studied by means of addition of specific activator or inhibitor of Wnt and RNAi. The study may be useful to elucidate the function of Wnt signal pathway in ovarian development, which may help to reveal the molecular mechanism for ovarian development regulation of Portunus trituberculatus.
三疣梭子蟹是我国重要的海洋经济动物之一,卵巢正常发育是其成功繁育的关键。到目前为止,三疣梭子蟹卵巢发育调控的分子机制尚不清楚。本课题组在前期研究中采用控制交配的方式阻止三疣梭子蟹卵巢发育,对成熟和未成熟卵巢进行比较研究发现,Wnt信号通路可能参与了卵巢的发育过程,但机制不清。本课题拟在前期研究的基础上,通过RACE技术结合EST序列获得Wnt信号通路部分关键组分基因的全长(比如Wntless、GSK3和Frizzled等)并分析三疣梭子蟹Wnt信号通路关键基因在卵巢发育不同阶段的表达特征;然后,分别采用RNA干扰技术、添加Wnt信号通路特异性激活剂或的抑制剂方法从正反两方面研究Wnt信号通路激活或者阻断对卵巢发育的影响。研究结果将阐明Wnt信号通路在三疣梭子蟹卵巢发育中的作用,有助于三疣梭子蟹卵巢发育调控的分子机制的解析。
