Transgenic watermelon (citrullus lanatus L.) plants were regenerated by inoculating cotyledon explants with agrobacterium tumefaciens stain LBA4404 harboring the binary vector pBin19 that carried the tobacco (Nicotiana tabacum cv Samsun NIV) modified class I chitinase and β-1, 3-glucannse genes. Transformants were selected for growth cpacity on medium with 75mg/l kanamycin. Molecular characterization by polymerase chain reaction and southern blot analyses confirmed the plresence of both the transgenes (chitinase gene, β-1, 3-glucannse genes) in the selected regenerants. Transgene expression was estimated by immunoblottin analysis. Six independent transgenic lines were recovered from a total of 350 inoculated explants, thus translating to a stransformation efficiency of 1.71%.. FA-resistant watermelon plants also were obtained using a watermelon tissue culture system. The putative musant were analyzed using AFLPsilver staining. And it was confirmed that when using primer M2E1, the obtained PCR products composed of a specific DNA band of FA-resistant watermelon plants.
