The early-bolting of carrot is a serious problem which influence the high efficiency and safe development of summer carrot's industrial product in Gansu, Qinghai and Neimeng plateau areas. According to the recent reports,it still remains unknown how endogenous hormone control the early-bolting of carrot.This project will explore the problem. Firstly, by using early-bolting and later- bolting carrot varieties, in summer season on plateau areas, the project is to study the regularity of the carrot's flower bud differentiation and early-bolting through plant anatomy ways. Secondly, by using ELISA and HPLC-MS/MS, the endogenous hormone contents of GA3, GA4, IAA, ABA and ZR are to be measured during the carrot's flower bud differentiation and early-bolting proper stages.Thirdly, the controlling effects of endogenous hormone on the carrot's flower bud differentiation and early-bolting are to be set forth by analyzing the relation between the content changes of GA3, GA4, IAA, ABA, ZR and the speed of carrot's flower bud differentiation, index of the flower bud differentiation and its velocity changes , the velocity of carrot's early-bolting and bolting stem growth, as well as the activity changes of source or sink and so on. Lastly, the controlling theory of endogenous hormone on carrot's early-bolting is put forward by validated tests from autumn to winter. Eventually,a reference theory is to be obtained for the study on a new later-bolting variety breeding and the safe and high effciency cultural technology of the carrot in summer season on plateau.
先期抽薹现象是影响甘肃、青海和内蒙等省(区)的高原夏季胡萝卜产业高效安全发展的突出问题。目前相关研究报道不能阐明内源激素对高原夏季胡萝卜先期抽薹的调控作用。本项目以易、耐抽薹胡萝卜品种为材料,在观察分析高原夏季胡萝卜花芽分化与先期抽薹发育规律的基础上,采用酶联免疫(ELISA)与液相色谱-串联质谱(HPLC-IT-MS/MS)相结合的方法,在花芽分化与先期抽薹过程中,适期测定高原夏季胡萝卜的内源GA3、GA4、IAA、ABA和ZR含量,研究分析内源激素含量变化与高原夏季胡萝卜花芽分化速率、花芽分化指数及其变化率,先期抽薹速率、薹茎伸长速率、抽薹指数及其变化率、源库活性等的关系,阐明内源激素对高原夏季胡萝卜花芽分化和先期抽薹的调控作用;并通过秋冬季验证试验,研究提出内源激素对高原夏季胡萝卜先期抽薹的调控理论,旨在为高原夏季胡萝卜耐抽薹新品种选育与高效安全栽培技术研究提供参考理论。
