The molecular mechanism on learning and memory is more and more attracting attention nowadays.The marginal division (MrD) in the brain striatum and it's learning and memory function were discovered by us.We have done a serious studies of neural biology, pathology and the behavior studies to investigate the structure and function of MrD. The results of these studies were confirmed by neuroscientists in the world. Some memory related genes were found in the prefrontal cortex, striatum, hippocampus and amygdala etc. But the expression and mechanism of the memory related specific genes of MrD are still a blank field.In this study, the genechip microarray analysis, qRT-PCR, Northern blotting and immunohistology combind with behavior methods were used to explore and compare the memory related gene expression profile of MrD of the striatum, striatum, prefrontal cortex, hippocampus and amygdala.In order to identify the memory relate gene exprssion programming in MrD,and the genotypic differences between MrD and other memory relatred brain areas. The specific molecular basis of different types of learning and memory genes in MrD would be verified in this study.The results of this study will provide new hint of diagnosis and therapy on dementia and other related diseases.It's possible to offer a new scientific design on the programme to enhance the learning and memory ability of human being.
目前学习和记忆的分子机理研究越来越受重视。我们发现了脑纹状体边缘区及其学习记忆功能,在神经生物学、病理学和行为学等多方面进行了深入的研究,发现其结构和功能特点都与纹状体其他部分以及其他记忆相关脑区不尽相同。其成果被国内外学术界公认。纹状体的学习记忆有关基因已有报道,但纹状体边缘区与学习记忆相关的特异性基因表达及作用机理的研究还是空白领域。本课题拟应用全基因表达组扫描技术,分析纹状体、边缘区、前额叶皮层、海马及杏仁核等与学习记忆相关脑区的基因及其表达状态即基因表达谱,然后应用荧光定量PCR、Northern blotting 和免疫组化方法在不同层面上验证。确认纹状体边缘区中与学习记忆有关的基因及其表达状态,获得区别于其他脑组织的与学习记忆主要相关的基因差异,确认脑纹状体边缘区对不同学习记忆类型的独特分子基础,从而为临床诊治智障及其相关疾病提供新线索,并可能为设计增强学习记忆的方案提供新思路
研究了纹状体边缘区的学习记忆的分子机理,并比较了边缘区和海马学习记忆功能的异同及其分子机理。
