LASS2/TMSG-1(homo sapiens longevity assurance homologue 2 of yeast LAG1/ tumor metastasis suppressor gene-1) is a new tumor metastasis suppressor gene and closely related with the invasion and metastasis of tumor. This research project intends to study the biological functions of LASS2/TMSG-1 gene through gene silencing, gene transferring, and co-immunoprecipitation method; discuss the regulation mechanism of V-ATPase (vacuolar H+-ATPase) on the LASS2/TMSG-1 gene; and look for the possible bridge protein between the LASS2/TMSG-1 gene and the V-ATPase by using gene chips and proteomics methods. Thus we will have a more systematic, comprehensive study on the anti-tumor mechanism of LASS2/TMSG-1 gene. This will help us to find the genes and therapeutic targets related to the metastasis of lung cancer and to provide a theoretical basis for the gene therapy for such diseases in Inner Mongolia region. For the first time, in particular, this project will apply LASS2/TMSG-1 to detect a large number of clinical specimens, and this will provide a large number of clinical and pathological basis for judging the prognosis of tumors and their growth, invasion and metastasis.
LASS2/TMSG-1基因是一种新的肿瘤转移抑制基因,与肿瘤的侵袭和转移密切相关。本课题拟在前期研究基础上,通过基因沉默、基因导入、免疫共沉淀等方法来进一步深入研究LASS2/TMSG-1基因的生物学功能,探讨V-ATPase(液泡型ATP酶)对LASS2/TMSG-1基因的调控机制;并利用基因芯片及蛋白组学方法来寻找LASS2/TMSG-1基因与V-ATPase之间的可能桥接蛋白,从而更系统、全面地研究了LASS2/TMSG-1基因的抗肿瘤机制,进而寻找与内蒙地区肺癌转移相关的基因和治疗靶点,为内蒙肺癌的基因治疗提供理论依据。尤其是本课题将LASS2/TMSG-1首次应用到了大量临床标本中的检测,从而为判断肿瘤预后及生长、侵袭转移提供了大量的临床病理依据。
LASS2/TMSG-1基因是一种新的肿瘤转移抑制基因,与肿瘤的侵袭和转移密切相关。与肺癌的关系研究甚少。本课题在前期研究基础上,通过基因沉默、基因导入等方法发现LASS2/TMSG-1基因与人肺癌侵袭迁移密切相关,初步认为人肺癌中V-ATPase(液泡型ATP酶)对LASS2/TMSG-1基因具有调控作用;并利用基因芯片方法来寻找LASS2/TMSG-1基因与V-ATPase之间的可能桥接蛋白,发现PIK3R5、AKT3蛋白可能参与了LASS2/TMSG-1基因在人肺癌侵袭转移的过程。本课题还将LASS2/TMSG-1首次应用到了大量临床人肺癌标本中的检测,发现LASS2/TMSG-1与肺癌的预后及生长、侵袭转移密切相关。
