Viral myocarditis (VMC), known as one of the most common cardiovascular diseases, is a serious threat to people's lives and health, and causes a high rate of sudden unexpected death. Problem is that some atypical death cases caused by VMC have no clear clinical symptoms and histopathological manifestation as well as comprehensive examination records, and thus an objective and detective indicator is necessary and demanding. We through genetic chip screening and genetic information found that microRNA-20 in VMC significantly changed and most likely ZFP148 was regulated by it. Based on our many years of research in this subject caused by sudden death, we carried out the following studies through the establishment of VMC model and primary myocardial cell culture: (1) we try to test the microRNA-20 and ZFP148, and explore their express specificity and regularity in myocardial tissue; (2) by interfering with intracellular microRNA-20 expression in myocardial cell and found that microRNA-20 regulate ZFP148 from both transcription and translation levels. The results of the research provide a new scientific, objective detection of target in forensic cases of sudden death caused by VMC, a new test targets for the clinical diagnosis of, but also it will help enrich our cardiovascular physiology and pathophysiology and understand of the mechanism of sudden cardiac death caused by VMC.
病毒性心肌炎(viral myocarditis,VMC)是目前严重威胁生命健康的最常见的心血管疾病之一,但在非典型VMC致猝死案件中,常出现病人临床症状不明显,缺乏临床资料,组织病理学表现不明显等矛盾,故非常有必要开发客观、可信的检测手段。我们通过基因筛查及遗传信息分析发现miRNA-20在VMC中有显著变化并极有可能对ZFP148基因有调控作用。因此,课题利用我们多年研究VMC致猝死的经验,通过建立VMC模型和原代心肌细胞培养模型等开展以下研究:(1)研究miRNA-20 及ZFP148在VMC心肌组织中表达的特异性及变化规律;(2)通过RNA干扰心肌细胞内miRNA-20表达,探讨miRNA-20对ZFP148基因在转录和翻译水平的调控作用。研究成果将为VMC所致猝死的法医学案件鉴定提供客观、敏感的依据,也将为VMC的临床早期诊断提供新的检测指标,并有助于探讨VMC致心源性猝死机制。
