Functional gastrointestinal disorders is a common complication after operation. Acupuncture is one of the hot spots in the treatment of gastrointestinal dysfunction. Our previous clinical studies have shown that electroacupuncture Zusanli can improve gastrointestinal dysfunction in patients with colorectal cancer after surgery. Transient Receptor Potential Vanilloid subfamily 1 (TRPV1) is a ligand-gated nonselective cation channel. Our previous study found that electroacupuncture Zusanli can significantly affect TRPV1 mRNA and protein expression. Further study found that TRPV1 expression is associated with gastrointestinal motility. But the exact mechanism is not clear. Recent studies show that activation TRPV1 can release substance P and combine with NK1 receptor in gastrointestinal smooth muscle, regulating gastrointestinal motility. In this study we try to use RT-PCR and Western blot and other biology techniques, to verify the signal pathway of acupuncture mediated by TRPV1 way, and clarify the mechanism of acupuncture regulating gastrointestinal smooth muscle contraction, providing a new choice for gastrointestinal dysfunction after operation.
术后胃肠运动功能障碍是术后常见的并发症。针灸是治疗胃肠功能障碍研究的热点之一。我们前期临床研究显示,电针刺足三里可以改善结直肠癌患者术后胃肠功能障碍。瞬时受体电位香草酸亚型1(Transient Receptor Potential Vanilloid subfamily 1,TRPV1)是一种配体门控的非选择性阳离子通道。我们前期研究发现,针刺足三里能够显著影响TRPV1 mRNA和蛋白的表达。进一步研究发现TRPV1表达与胃肠运动相关。但确切机制尚未清楚。最近研究表明激活TRPV1可以释放P物质与胃肠平滑肌上NK1受体结合,调节胃肠功能。本课题拟通过RT-PCR和Western blot等分子生物学技术,进一步验证针刺足三里通过TRPV1介导的信号通路的方式和途径,并阐明针刺足三里调节胃肠平滑肌收缩的机理,为术后胃肠功能障碍的治疗提供新的思路。
