The applicant previously work has shown that baculovirus (Antheraea pernyi nuclear polyhedrosis virus [ApNPV]) is capable of stimulating innate immune response of avian both in vitro and in vivo. In order to further study the mechanisms of baculovirus-induced activation in avian immune cells, GST-Pull Down, DNA enzyme Ⅰ footprinting analysis, RNAi,magnetic beads, spectrometry and N-terminal sequencing combined with bioinformatics and preliminary research results were adopted to investigate the membrane receptor and the targets in the baculovirus-activated immune cells. Further, baculovirus DNA sequences which were interacted with the target receptor were also analyzed. To clarify the membrane receptor by which the baculovirus enter the cell and the target protein on which the virus affect, will in theoryly reveal the molecular mechanisms and the signal transduction pathways of the baculovirus-induced immune cells in avian. The identification of the underlying regulatory mechanisms of baculovirus-induced avian immune cells activation provides an important basis for the development of new immunomodulators and vaccine adjuvants for better treatment of avian diseases and protect the human health.
申请人前期研究结果表明柞蚕杆状病毒在体内和体外都能有效激活禽类的天然免疫。为进一步研究杆状病毒激活禽类免疫细胞的作用机制,本项目拟采用GST-Pull Down、DNA酶Ⅰ足迹分析、RNAi、磁珠亲和富集、质谱和N-端测序分析等实验技术与方法,结合生物信息学和前期研究成果,对柞蚕杆状病毒激活禽类天然免疫系统过程中病毒作用于禽类免疫细胞的膜受体、细胞内的作用靶点及与靶点受体相互作用的杆状病毒DNA序列特征等问题进行研究,阐明杆状病毒通过何种膜受体进入细胞内部、作用于何种靶蛋白,从而激活天然免疫,可望在理论上揭示柞蚕杆状病毒激活禽类天然免疫的分子机制,建立柞蚕杆状病毒激活禽类天然免疫的信号转导途径。该项目的完成对探索作用强,起效快的新型疫苗或免疫调节剂,防治禽类传染病及保障人类健康具有重要意义。
