This project is a continuation of the Youth Fund of National Natural Science Foundation project "Oil Crops Endogenous Reference Gene Research". For the practical problems of genomic DNA fragmentation, degradation extraction and detection difficulties of deep processing of agricultural products encountered in the original project implementation as well as routine testing work, this project is scheduled to be resolve the above questions. Contents of this project, include research and establish suitable efficient extraction method for the small pieces of genome DNA from deep processing agricultural products; improvement of the existing PCR detection methods, shortened needed detection target length, establish suitable ultra-short target detection method for small fragment of genomic DNA extracted from deep processing agricultural products; combination of the ultra-short target detection methods and digital PCR technology, to establish the mathematical model of the broken DNA length distribution based on the statistical analysis of separation of targets in a certain length; analysis of different deep-processed products genomic DNA length distribution based on the broken DNA length distribution model; analysis of the degradation dynamics of genomic DNA in different processing; the establishment of a technical system for the precise identification of the raw materials in deep processing agricultural products and to contribute to the food safety of the people.
本项目是国家自然科学基金青年基金项目"油料作物内参照基因研究"的延续。针对原项目执行以及日常检测工作中遇到的深加工农产品中基因组DNA断裂、降解,以及由此导致的提取和检测困难的实际问题,本项目计划系统研究并建立适合深加工农产品的小片段基因组DNA高效提取方法;改良现有PCR检测方法,缩短需要的检测靶标长度,建立适合深加工农产品中提取的小片段基因组DNA的超短靶标检测方法;结合超短靶标检测方法和数字PCR技术,以一定长度间靶标的连锁和分离为基础,进行统计学分析,建立破碎DNA长度分布的数学模型;利用破碎DNA长度分布模型分析不同深加工产品中基因组DNA长度的分布;分析不同加工工艺过程中基因组DNA的降解动态;最终建立适合深加工农产品原材料精准鉴定的技术体系,为人民群众的食品安全做出贡献。
本项目是国家自然科学基金青年基金项目“油料作物内参照基因研究”的延续。.针对日常检测工作中遇到的深加工农产品中基因组DNA断裂、降解,以及由此导致的提取和检测困难的实际问题,本项目开展了系统研究。.针对加工造成的DNA降解和破碎难题,本项目建立了适合深加工农产品的系列小片段基因组DNA高效提取方法;针对深加工产品中含有的干扰物和小分子DNA难题,本项目比较了现有方法的优缺点,并建立了高效准确的基因组DNA含量测定方法;针对DNA片段断裂问题,本项目开发了全新的PCR检测方法,缩短需要的检测靶标长度,建立更适合深加工农产品中提取的小片段基因组DNA的超短靶标检测方法;针对收储加工企业的实际需要,本项目开发了适合降解蛋白质的现场快速检测方法;并对深加工产品中外源蛋白为代表的有效成分的降解过程进行了跟踪检测。.总体上,本项目推进了适合深加工农产品原材料精准鉴定的技术体系的建立,完成了预期任务。
