Nano-TiO2/Nano-ZnO nano-composites can be altogether used in these fields of medicine, cosmetics, textiles, and energy materials. Especially,more attractions have been focused on their new functional developments of antiseptic bionic nano-skeletal repair materials and anti-cancer target nano-medicine carriers. Although some papers have reported their individual toxicity, almost none on the characteristics and mechanism of their combined toxicity has been investigated. As classical cancerous and non-cancerous models, both human liver cancer cell HepG2 and human liver normal cell HL-7702 can scientifically evaluate the combined toxicity of Nano-TiO2/Nano-ZnO and accurately screen target nano-drug based on their specificity. The project aims to focus on the combined toxicity and their synergism in the aspects from biochemistry and molecular biology. This study will confirm whether Nano-TiO2/Nano-ZnO is safely applied for the transplantation of human bone, the manufacture of sun-screen, the development of the target anti-cancer nano-drug vector, as well as to broaden the cognitive concept of public health and environmental toxicology.
Nano-TiO2/Nano-ZnO纳米复合材料常被共用于医药卫生、化妆品、纺织及能源材料等领域。尤其作为新型抗腐仿生骨骼纳米修复材料和抗肿瘤药物靶向纳米载体深受关注。尽管它们各自单独纳米毒性有一些报道,但它们组合对人体细胞毒性特征及机制至今了解甚少。而人肝癌细胞HepG2和人肝正常细胞HL-7702作为一组经典已癌化和未癌化对比细胞模型能更加科学性判断Nano-TiO2/Nano-ZnO对人体细胞组合毒性及根据其特异性准确筛选靶向药物。本项目拟重点研究Nano-TiO2/Nano-ZnO细胞组合毒性及从生物化学和分子生物学多角度探索其组合毒性分子机制。此研究有助于确定Nano-TiO2/Nano-ZnO是否安全应用于人体骨骼移植、防晒霜研制和靶向纳米药物载体开发等方面并拓宽公共卫生及环境毒理学认识范畴。
