Flaxseed cake resource, which contains a variety of biological function components as by-product of flaxseed oil, faces the problem of low utilization during the oil production. Based on our previous works of extracting compound information from instruments of chromatography-high resolution mass spectrometry, this project aims to take full advantage of untargeted metabolomics and proteomics coupled with advanced methods of Chemometrics to screen key antioxidants of endogenous metabolites and peptides from protein of flaxseed cake, and, meanwhile, study the variation law of these components during the stir-frying procedure of flaxseed. The research will not only explain the quantitative relationship between oxidant capability of flaxseed cake and the contents of antioxidant components, but also provide efficient methods for rapidly characterizing the quality of flaxseed cake in the stir-frying procedure. To the best of our knowledge, this ideology has not been reported before. The implementation of this project could determine clearly the key antioxidant metabolites and peptides in the flaxseed cake and their contribution to the quality of flaxseed. Additionally, the research results might improve the high additional-value of industrial products related to the flaxseed cake resource, and could be helpful for its antioxidant theory development.
针对胡麻炒籽榨油副产物饼粕富含多种生物活性成分,但资源利用率低这一问题,项目拟在前期色谱联用高分辨质谱实现植物样本中化学成分剖析研究的基础上,拟采用针对内源性代谢物的非靶向代谢组学技术以及针对蛋白质分解后活性肽段的蛋白质组学技术,结合化学计量学物质信息智能化剖析新方法,筛选胡麻籽饼粕中具有抗氧化活性的蛋白质分解肽段及内源性代谢物成分,系统研究这些关键抗氧化活性成分构成及含量在胡麻籽炒制过程中的变化规律,阐明胡麻籽饼粕中蛋白质活性肽段、内源性代谢物组分与宏观抗氧化能力之间的构效关系,基于抗氧化活性成分变化建立精准表征胡麻籽饼粕宏观品质的化学计量学预测模型。研究思路未见文献报道。本项目的实施不仅会明确胡麻籽饼粕中具有抗氧化活性的关键内源性代谢物、蛋白质活性肽段及其与胡麻籽饼粕品质之间的关系,促进胡麻籽饼粕资源的高工业附加值、精细化工产品的研发,还将丰富胡麻籽饼粕的抗氧化理论研究。
本项目针对胡麻炒籽榨油副产物饼粕富含多种生物活性成分,但资源利用率低这一问题,利用色谱联用高分辨质谱的组学技术,发展一系列化学计量学物质信息智能化剖析新方法,剖析了胡麻籽饼粕中内源性物质构成、含量及比例关系,将不同阶段的胡麻籽饼粕与DPPH等体外抗氧化结果进行结合,利用化学计量学方法筛选了具有抗氧化活性的化合物,并开发了活性成分筛选与确定交互平台AntDAS[O]。并根据宁夏地区胡麻籽企业中典型的热加工工艺,设计了胡麻籽炒籽榨油实验,对化学物质在热加工过程中的变化开展了系统性研究,明确了化合物在炒制过程中的变化规律。基于现代分析技术,构建了评估胡麻籽饼粕品质的化学计量学模型。本项目为我国西北地区胡麻籽饼粕资源的精深加工利用,促进胡麻籽饼粕资源的高工业附加值、精细化工产品的研发提供了科学支持。
