This project consists of two parts: Studies on 3-D structure-function of Earthworm Fibrinolytic Enzyme(I) and phospholipase A2(2)..The 3-D structure of Earthworm Fibrinolytic Enzyme A(EFEa), a protein which functions both as a direct fibrinolytic enzyme and a plosminogen activator, has been determined by MIR method and refined to 2.3? resolution. It is the first of an earthwonm fibrinolytic enzyme component and of a serine protease origination from the annelid worms. It provides a lead template for accurately modeling other homologaus family members; The venom from Ophiophagus Hannah (King Cobra) contains two acidic phospholipase A2 (PLA2). The crystal structures of these two enzymes were determined by X-ray crystallography. The PLA2-I structure is the first structure of snake venom PLA2 that contains the pancreatic loop and the first structure of a PLA2-type cardiotoxin. The protuberance of the pancreatic loop on the interface recognition sites plane may affect the binding of the aggregated substrates to the enzyme, thus decreasing the enzymatic activity for this PLA2 compared with PLA2-II. In addition, crystal structure of an acidic PLA2 from the venom of Naja Kaouthia was also determined in two crystal forms. The results emphasize the physiological importance of venom PLA2 oligomer state.
