As one of the most important plant hormone, auxin regulates plant development and differentiation all the way. Till 1997, Tim Ulmasov's group cloned the first auxin response transcriptional factor, ARF1 in arabidopsis. Sometime later, this group got some other ARF family genes by cDNA library screening. Few ARF family genes were cloned in grass family, except for maize VP1. Due to the difference between dicot and monocot plant, it's agricultural and theoretical important to study the auxin regulation mechanism in monocot plant, especially in rice. In this proposal, a rice ARF family gene rARF1, both the cDNA and the genomic coding region was cloned. Some basic functional research was done. Furthermore, we set up an e-cloning method, based on rice full genomic sequence and homology between species and obtained some important full length cDNA of new rice genes.
ARF1是涉及植物生长素信号转导路线中同后期靶基因的表达调控有关的一个重要的转录因子,与Aux/IAA形成二聚体,处于信号转导的中心位置.采用差减cDNA技术已从水导幼穗中获得与ARF1高同源的cDNA克隆,将从已建立的水稻幼穗全长cDNA库中分离全长rARF1基因并完成序列分析.以全长rARF1 cDNA转化番茄进行异位表达研究其功能.
