Electrospray-Time of Flight Mass Spectrometry (ESI-TOFMS) is the kew scientific instrument in Life Science and Chemistry, which has a great impact to the frontier of the whole science. As well known, USA scientist, Dr. Fenn wins the 2002 Nobel Prize of Chemistry for his invaluable contribution in innovation of ESI-MS.By the end of this project, we have set up an first home-made ESI-TOFMS instrument in China; we have studied the pulsed-ESI for the first time and investigated the bio-ion behavior under pulsed conditions; and we have hyphenated micro-FIA with the ESI-TOFMS.(1)The physical and Chemical process of Bio-molecules under the pulsed-ESI condition has been investigated. Under pulsed condition, the ions generated from ESI can be fully utilized by MS, with the advantages of enhanced pulse intensity of ion signal and reduced sample flow rate in average. This technique is quite applicable to the micro-CE/HPLC/FIA in protein separation and identification. (2)The homogeneous ion-optics has been studied and used in our instrument. Under homogeneous conditions, the wide-ion beam can be relatively focused in compensation with the ion energy dispersion and ion space dispersion. Therefore, the resolution can be improved tremendously, which benefits to the large bio-molecules. With all studies, we have optimized the ion optical system for both ion extraction and acceleration.(3)We have set-up our ESI-TOFMS with an orthogonal ion acceleration mode and a reflectron mode, and optimized all the electronic parameters. We have tested our instrument with various bio-samples, showing that the mass resolution has reached 13000, and the sensitivity can be as high as 10 pg/ml resinpine with a S/N ratio of >100..The success in building of home-made ESI-TOFMS can be highly evaluated for its significances in Bio-science in China, and also in the life and chemical instrument industry. It will promote our own ESI-MS products in the near future with the advance technologies in the world-front levels.
