。The completed genomic sequencing on pigs conduces to the further study on pigs' heredity, seed breeding and nutrition regulation at the molecular level. Neuropeptides, the significant signaling molecules in the system of neuroendocrine, have been improved to play an influential role in animals' such vital movements as growth, metabolism, and multiplication. At present the function studies of some neuropeptides on livestock and poultry have been reported. But these studies, which were experimented on one single gene or several genes,have not systemically revealed pigs' unknown neuropeptides and roundly explored the regulation of the neuropeptides on given animals. Based on the pigs' genomic information resource, this study utilizes the real-time PCR to research the expression characteristics of Rongchang pigs. And by using the bioinformation analytical techniques we carry out the peptides analysis on high-abundanced tissues and resolve the quantity and structures of Rongchang pigs' mature neuropeptides. Comparing the neuropeptides' expression differences and the function of the different genes in the growth and development of Rongchang pigs and Landrace pigs, we reveal the regulating hereditary traits of Rongchang pigs' neuroendocrine, and enrich the genetic resources of Rongchang pigs', which can ground the study of Rongchang pigs well in the mechanism of growth and metabolism, nutritional requirements and the regulation of the products quality.
猪基因组测序的完成,有助于提升在猪遗传育种和营养调控分子水平的研究。神经内分泌系统的重要信号分子-神经肽被证实在动物的生长、代谢及繁殖等生命活动中发挥了举足轻重的作用。目前,部分神经肽在畜禽动物上的功能研究已有相关报道,但仅针对单个或几个基因展开了研究,尚未系统地挖掘未知的猪神经肽基因和全面地探究神经肽在特定品种中的调控作用。本项目基于猪基因组信息资源,利用生物信息分析技术,全面而系统地筛查猪神经肽基因,并进一步利用荧光定量PCR技术考察优良地方猪种-荣昌猪神经肽基因的组织表达特征,并对高量表达的组织进行肽组学分析,解析荣昌猪成熟神经肽的数量及结构;并通过考察荣昌猪与长白猪神经肽基因在生长发育过程中各组织的转录表达差异,开展差异基因的功能研究,解析两品种猪不同生长、生产及繁殖的神经内分泌调控机制,进一步丰富荣昌猪的遗传资源,为荣昌猪的生长代谢、营养需要及产品质量的调控奠定坚实的基础。
神经内分泌系统的重要信号分子-神经肽被证实在动物的生长、代谢及繁殖等生命活动中发挥了重要的调控作用。目前,部分神经肽在畜禽动物上的功能研究已有相关报道,但仅针对单个或几个基因展开了研究,尚未系统地挖掘未知的猪神经肽基因和全面地探究神经肽在特定品种中的调控作用。本项目基于猪基因组信息资源,利用生物信息分析技术,全面而系统地筛查得到了116个猪神经肽基因,预测了459个成熟神经肽,并进一步利用荧光定量PCR技术考察了这些神经肽基因在优良地方猪种-荣昌猪神经肽基因的组织表达特征,并对高量表达的组织-脑组织进行了组学的分析,证实了75个表达的神经肽基因。并通过考察荣昌猪与外种猪神经肽基因在生长发育过程中各组织的转录表达差异,获得了差异表达基因,一定程度地解析了荣昌猪优良地方猪种和外种猪不同生长、生产及繁殖的神经内分泌调控机制。我们的研究不仅丰富荣昌猪的遗传资源,也为荣昌猪的生长代谢、营养需要及产品质量的调控奠定了坚实的基础。
