Volatile organic compounds (VOCs) biological sample (exhaled breath,blood and urine) analysis is a novel approach that has recently been developed for the screening and diagnosis of diseases. Because this approach exhibits the advantages of being convenient and non-invasive, VOCs analysis has drawn increasing attention from researchers. Many recent studies have demonstrated the connection between the specific VOCs and various forms of cancer, including lung cancer and breast cancer. However, these findings are complex, can not be compared, mainly due to the lack of standardization subjects. In this study, we want through in vitro and in vivo experiments, by using biomedical technology (flow cytometry, confocal microscopy), analytical chemistry techniques (solid phase micro extraction, gas chromatography - mass spectrometry analysis) and metabolomics methods (fingerprinting analysis of volatile organic compounds) to establish a standardized volatile biomarkers screening platform. Comparative fingerprinting analysis of volatile organic compounds were conducted during the growth, necrosis, apoptosis of tumor cells; tumor resection, tumor growth, necrosis and apoptosis. Specifically volatile biomarkers were founded, and this may helpful for clinical cancer diagnosis and course of judgment.
挥发性有机化合物生物样本(呼出气、血、尿样本)分析是近年来发展起来的一种新的筛查,诊断疾病的方法,由于其便捷,无创,患者耐受性好等优点,越来越引起研究者的重视。已有研究证实不同癌症有着其特异性的挥发性代谢物,如肺癌、 乳腺癌、结直肠癌、胃癌等。但这些研究结果各异,不能进行比较,主要原因为受试对象缺乏标准化。本研究拟通过离体组织培养和在体实验,利用生物医学技术(流式细胞仪、激光共聚焦显微镜),分析化学技术(固相微萃取、气相色谱-质谱联用分析技术)和代谢组学方法(挥发性有机化合物指纹谱分析)建立一个标准化的挥发性生物标记物筛选平台。通过对肿瘤细胞生长、坏死、凋亡、肿瘤组织切除、肿瘤组织生长、坏死、凋亡等情况下进行挥发性有机化合物指纹谱比较分析,筛选出明确的癌细胞生长、凋亡和坏死的挥发性生物标记物,为临床癌症诊断及病程判断提供依据。
