Starch is the major component in maize kernels, providing staple carbohydrate source for humans and livestock as well as raw materials for biofuel industry. Hence, improving maize kernel starch content will not only has the potential to increase overall yields, but also help meet industry demands. Previously, a major QTL for starch content on chromosome 3, qHS3, was identified with an F2:3 population derived from B73 with low-starch content and DHLoPro1 with high-starch content. To fine map and clone the QTL, near-isogenic lines (NILs), in which DHLoPro1 as donor parent and B73 as recurrent parent, were developed on that basis. Association analysis, over-expression and knock out of candidate gene to explore gene function will help further explain starch biosynthesis and accumulation in maize kernels, which provide a guide of improving starch content in maize kernels by MAS.
淀粉作为玉米籽粒最主要的组成部分,不仅为人类和动物提供重要的能量来源,还为各种工业生产加工提供原料。因此,提高玉米籽粒淀粉含量不仅能增加玉米产量,还能满足工业需求。本研究以利用高淀粉自交系(DHLoPro1)和普通自交系(B73)组配的F2:3群体在3号染色体上定位到一个控制玉米籽粒淀粉含量的主效QTL(qHS3)为基础,拟构建以B73为轮回亲本、DHLoPro1为供体的近等基因系,对该QTL进行精细定位和图位克隆,并利用候选基因关联分析、超表达、基因敲除等方法研究该基因的功能,初步阐明该QTL调控玉米籽粒淀粉合成和积累的遗传机理,并为通过分子育种改良玉米籽粒淀粉含量提供理论指导。
