Hemorrhagic fever with renal syndrome(HFRS)is a worldly distributed viral disease with high incidence and harmfulness.There have no special effective treatment. The key point obstructing the stuy of pathogenesis and antiviral drugs is lack of proper animal infectious model.To determin it,we construct a cell infectious model to determine the enzyme fuction and activity of L protein confirmly and quantitatively, which can be applied to screen antiviral drugs.Firstly,construct dual report gene expres plasmid containing lateral sequence of S segment,then transfect it to Vero-E6, finally, it was infected with hantaan virus. The study is innovative,the transfection rate of constructed system is much high than double plamids without control.
构建一个能将报告基因可调控性转录仅被HFRS病毒L蛋白识别并翻译的单链RNA的细胞感染模型,能简便地定量测定病毒L蛋白的活性,反映病毒在真核细胞内的转录及翻译水平,并应么四P蜕秆∫圆《綥蛋白为作用靶点的高效抗病毒药物,并可分析其作用环节。解决目前HFRS病毒感染中治疗、药物筛选及分子病毒学研究中缺乏合适感染模型的关键问题。
