In human, variants of CISH are associated with susceptibility to diseases caused by diverse infectious pathogens,however the functions of porcine CISH remain elusive. In the present study, we will perform the cloning, subcellular localization and antibody preparation of the porcine CISH, identify the promoter region and the reaction region of LPS, on these basis, analysis the role of porcine CISH in regulating TLR4/NF-κB signal pathway by gene over-expression and RNAi, analysis the role of CISH and TLR4/NF-κB signal pathway on diarrhea in weaning pigs between Minzhu and Landrace breeds. This research can elucidate the molecular mechanism of porcine CISH in regulation TLR4/NF-κB signaling pathway,contribute new candidate genes and ideas for for the study on disease resistance in pig breeding, and also provide theoretical basis for the breed conservation and utilization in livestock production of Minzhu pigs.
CISH基因的遗传变异与人类多种传染性疾病显著相关,但是猪CISH基因功能未知。本研究拟进行猪CISH的克隆、亚细胞定位和抗体制备;鉴定CISH启动子区域及其LPS反应区域;在此基础上,利用基因过表达和RNAi技术分析猪CISH在TLR4/NF-κB信号通路中的作用;利用Real-time PCR技术分析CISH及TLR4/NF-κB信号通路基因在民猪和长白猪断奶腹泻中的作用。本研究将阐明CISH调控TLR4/NF-κB信号通路的分子机制,为抗病育种研究提供新的基因资源和研究思路,为民猪的保种及其在生产中的合理利用提供新的理论依据。
CISH基因的遗传变异与人类多种传染性疾病显著相关,但是猪CISH基因功能未知。本研究首先进行猪CISH的克隆、亚细胞定位、抗体制备和启动子区域鉴定;在此基础上,研究猪CISH在TLR4/NF-κB信号通路中的作用;最后研究CISH及TLR4/NF-κB信号通路基因在细胞炎症反应中的作用及其与仔猪腹泻的相关性。研究发现CISH基因定位于细胞质,并获得CISH抗体;CISH及TLR4/NF-κB信号通路基因参与细胞炎症反应;CISH和IL-6基因遗传变异与民猪仔猪腹泻相关。本研究结果为猪抗病育种研究提供新的基因资源和研究思路,为民猪的保种及其在生产中的合理利用提供新的理论依据,具有一定的科学意义和应用前景。
