Methyl tert-Butyl Ether (MTBE) is a currently worldwide used fuel antiknock additive substituting for lead alkyls. Previous studies show that MTBE appeared to be positive in the carcinogenicity tests at high dose for a variety of species and target organs. TAME is another prosperous oxygenate additive. In this study, the cellar DNA/hemoglobin adductions with MTBE in male mice were investigated in a wide dose range by an advanced ultrasensitive technique, accelerator mass spectrometry (AMS). The decay kinetics of MTBE-DNA adducts after a single exposure was also exmined. It's our first finding that MTBE can bind to cellar DNA and hemoglobin in vivo increasingly with increasing dose in lg/lg presentations. The distribution of MTBE adducts in different issues is lung>liver>kidney> hemoglobin. Some MTBE-DNA adducts both in lung and liver can exist for quite a long time. Besides, we also studied the interactions of MTBE/TAME and hemoglobin(Hb)/bovine serum albumin(BSA) in vitro by FT-IR and 1H-NMR spectoscopies, and, the effects of MTBE/TAME on L-02,the normal human liver cells , which showed that TAME has more severe effection on both the growth rates and the shapes of L-02 than MTBE.
机动车汽油已大规模使用MTBE取代四乙基铅,但MTBE任有显著的毒性。国际上正开发新添加剂TAME以降低毒性,我们用加速器质谱法(AMS)研究极低剂量的TAME与动物体内DNA和血红蛋白的加和及其化学作用机制。建立极灵敏的生物标志物,并用紫外,核磁,荧光等化学谱学手段,体外研究TAME与生物大分子的化学作用机制。
