Oxidative stress is a major problem in the dairy farming industry, antioxidants can alleviate oxidative stress in dairy cattle. Health mammary cells of dairy cow are selected as the research object, tea polyphonals are selected as modulators of oxidative stress, to build oxidative injuried model using oxidative stressed mammary cells of dairy cows induced by H2O2, and observe the intervention effect of tea polyPhenols on oxidative damaged dairy cow mammary cell in this study. Primary oxidative damaged mammary cells are selected as the research object, to explore tea polyphenols whether activate Nrf2-ARE pathway to against oxidative stress by observing Nrf2 and p-Nrf2 protein expression changes of oxidative injuried mammary cell? to investigate Nrf2-ARE pathway by activating which expression of phase Ⅱ detoxifying enzymes to against oxidative stress by transfecting Nrf2 specific siRNA? And revealed that mechanisms of tea polyphenols protect mammary cells against oxidative injury. This study systemly illustrates effect of tea polyphenol on Nrf2-ARE signaling pathway from the animal tissue, cell and molecular level by cell culture, which has important significance to ensure that the dairy cows can maintain the normal function of lactation and improve production performance in peak lactation and during transition, and provide a theoretical basis for tea polyphenol as a natural antioxidant in livestock production applications.
氧化应激已成为困扰奶牛饲养的难题,利用抗氧化剂缓解奶牛氧化应激已成为当前研究热点。本研究拟以健康奶牛乳腺细胞为研究对象,茶多酚为氧化应激调控剂,采用H2O2处理构建奶牛乳腺细胞氧化损伤模型,阐明茶多酚对氧化损伤奶牛乳腺细胞的干预作用;然后以原代乳腺细胞氧化损伤模型为研究对象,通过观察氧化损伤乳腺细胞Nrf2、p-Nrf2蛋白表达量的变化,探讨茶多酚是否通过激活Nrf2-ARE通路对乳腺细胞发挥抗氧化作用?通过转染Nrf2特异性siRNA,探讨Nrf2-ARE通路是通过启动下游哪个第Ⅱ相解毒酶的表达来发挥抗氧化作用?进而揭示出茶多酚保护乳腺细胞对抗氧化损伤的作用机制。本研究系统地从动物组织、细胞和分子水平上探讨了茶多酚对Nrf2-ARE通路的影响,对于确保奶牛在泌乳高峰期和围产期内维持正常泌乳功能和提高生产性能具有重要意义,也可为茶多酚作为一种天然抗氧化剂在畜牧业生产中应用提供理论依据。
