Airway remodeling is the important pathophysiology foundation in the occurrence and development of chronic obstructive pulmonary disease(COPD).RhoA /ROCK signal pathway is closely related to the activation of neuroendocrine inflammatory factor of airway remodeling after COPD, and it is an important target for intervention of airway remodeling and the prevention and treatment of COPD. Traditional Chinese Medicine believes that the pathogenesis key of COPD is asthenia-phlegm-stasis.It is the theory that the asthenia of lung,spleen,kidney is the root and blood stasis and phlegm obstruction is the branch.Besides the method of tonifying qi and promoting blood circulation and resolving phlegm is the basic therapeutic methods of COPD.The previous findings showed, the method of tonifying qi and promoting blood circulation and resolving phlegm can reduce airway inflammation, regulate oxidant / antioxidant imbalance,and improve symptoms and lung function and reduce exacerbations.Based on the preceding research, this program will use research methods of combination of in vitro and vivo. Serum pharmacological methods such as ELISA ,Western blot, RT-qPCR and flow cytometry and other modern molecular biology techniques will be used to intensively study the relationship between the method of tonifying qi and promoting blood circulation and resolving phlegm on airway remodeling and Rho/ROCK signal pathway ,to find its target further revealing the biological foundation of the method of tonifying qi and promoting blood circulation and resolving phlegm for the prevention and treatment of COPD to provide theoretical evidence of its clinical application.
气道重塑是慢阻肺发生发展的重要病理学基础,气道重塑过程中神经内分泌细胞因子的激活与RhoA/ROCK信号通路关系密切,阻断RhoA/ROCK通路是阻抑气道重塑、防治慢阻肺的重要策略之一。中医认为,慢阻肺病机关键是"虚-瘀-痰",肺脾肾三脏亏虚为本,血瘀痰凝为标,益气活血化痰是其基本治法。既往研究表明,益气活血化痰法能够减轻气道炎症、调控氧化/抗氧化失衡,改善肺功能和症状,减少急性加重次数。本项目拟在前期基础上,采用体内外实验结合的方法,运用ELISA、Western blot、RT-qPCR、流式细胞术等分子生物学技术,以RhoA/ROCK信号通路为切入点,探究益气活血化痰法干预慢阻肺气道重塑的分子机制,为其临床运用提供理论依据。
