More and more evidences showed that the "scilent cells" in the normal or degenerated nucleus pulposus (NP) behave like proginiter cells or mesenchymal stem cells.To study how to activate these "scilent cells" to initiate endogenous repairing could provide a new insight into the biological therapy of degenerated intervertebral disc.It has been verified that cells derived from nucleus pulposus could be activated by exogenous stem cells,but the exact mechanisms have been kept unknown yet.With coculturing of exogenous stem cells and the "scilent cells"derived from the nucleus pulposus, the abnormal expression of any cytokines could be detected by Genechip teconology. This assay provide a possibility to discover the valuable activating cytokines and activating signal net.This study will focus on screening the valuable activating cytokines by collecting the NP-derived cells which are cocultured with the ectogenous stem cells,and exploring the potential activating mechanisms of the scilent cells.The outcomes may cue an accurate and efficient method for activating the "scilent cells"in the NP tissue..
有证据显示成人健康及退变髓核组织中均存在着功能"静默"状态的髓核基质干细胞。研究如何"激活"内源性"静默"细胞,从而启动内源性修复,将为椎间盘退变性疾病的生物学治疗提供新的思路。髓核来源细胞与多种干细胞共培养后被"激活(增殖及分泌基质能力增强)"的现象已被众多研究所证实,但此现象背后的机制目前尚不清楚。基因芯片技术可用于检测"静默"细胞与干细胞共培养后多种不同细胞因子的异常表达,从而为筛选有价值的激活因子和信号激活通路提供可能。本研究将采用自体脂肪干细胞与髓核"静默"细胞共培养的方式,利用基因芯片技术,筛选共培养后基因谱表达变化情况,分析可能的激活因子及"静默"细胞被激活信号通路,并进行初步验证,从而为进一步的机制研究提供科学依据。
