Moreover, the specific dynamin 1 inhibitor, dynasore, significantly reduced epileptic seizure in li-pilocarpine rats and epileptic discharge on the epileptic hippocampus slices. Dynamin 1 plays an important role in regulating synaptic vesicle recycling of endocytosis; unlike kiss-and-run and clathrin mediated endocytosis (CME) pathway, the activity - dependent bulk endocytosis (ADBE) is the dominant endocytosis pathway during the most main strong high frequency environment .This project intends to patients with drug-resistant epilepsy, animal models, in vitro brain slice model and cell model as the research object, on the basis of previous research results through gene point mutation, import the peptide inhibitors such as molecular biology technology, presynaptic imaging technology, multi channel record and patch clamp technique in vivo, clear dynamin 1 in epilepsy is mainly through ADBE pathways regulate synaptic vesicle recycling work; Looking for intervention dynamin 1 mediated ADBE way to influence the best effect of epileptic seizure link, explore dynamin 1 mediated ADBE way of related mechanism of epilepsy, in search of new drug targets
课题组研究发现,dynamin1的表达在癫痫动物和耐药性癫痫患者脑组织中显著增高,其特异性抑制剂dynasore明显减少了模型鼠癫痫发作及癫痫脑片中的痫样放电。现已证实dynamin1在调节突触囊泡再循环的内吞中起重要作用;它介导的活性依赖的批量囊泡内吞(activity-dependent bulk endocytosis,ADBE)途径是强力高频刺激环境下最主要的内吞途径。本项目拟以耐药性癫痫患者、动物模型、脑片模型及细胞模型为研究对象,在前期研究结果的基础上通过基因点突变、肽抑制剂等分子生物学技术、突触前成像技术、在体多通道记录及膜片钳技术,明确dynamin1在癫痫中是否通过ADBE途径调节突触囊泡循环发挥作用;寻找dynamin1介导的ADBE途径中影响癫痫发作的关键作用环节;探索与dynamin1介导的ADBE途径相关的癫痫发病机制,为寻找新的药物靶点提供理论依据。
中文摘要.癫痫发作涉及神经递质的快速释放与回收,这需要突触囊泡快速循环来维持。神经元活动增强时,需去磷酸化的dynamin1介导活动依赖性批量內吞(ADBE),提供足量的囊泡,以适应陡然增高的突触传递压力,来维持囊泡循环。(研究背景).重要研究结果.1、Dynamin 1蛋白及mRNA在癫痫动物模型及癫痫患者中表达增高,Dynasore在体内和体外模型中都能抑制痫样放电.2、Dynasore 对大鼠痫性发作的影响.匹鲁卡品造模后,20min、30min、40min、50min、60min 5 个时间点,10uM dynasore 组发作严重程度均低于对照;Dynamin 1 蛋白表达在对照 组无明显差异,10um dynasore 组dynamin 1 蛋白水平降低。.癫痫动物模型和癫痫患者中,dynamin 1的 mRNA 和蛋白水平均升高,特异性抑制剂 dynasore 在10uM时具有体内匹罗卡品/体外癫痫脑片模型抗癫痫作用,说明 dynamin 1 可能和癫痫发作相关。.3、干预前后dynamin1磷酸化水平在癫痫动物模型中的变化.3.1 Dynamin 1 总蛋白在各组表达无显著差异。Dynasore 100、160 μM 组海马的 dynamin 1 磷酸化水平显著增高。.3.2 10 μM和20 μM dynasore能抑制脑片痫样放电,动作电位的频率分别从1.302±0.617 / 2.314±1.437 Hz减少到0.743±0.519 / 1.217±0.926 Hz。.3.3 10umol dynasore降低mEPSCS的频率,增加mIPSCS频率,对二者幅值均无影响;此外,该浓度dynasore降低sIPSCS的频率,对幅值无影响。.4、Dynamin 1抑制剂dynasore对癫痫细胞的影响.4.1 20uM dyansore
