Acute pancreatitis (AP) is one of common acute abdominal diseases. Qingyi decoction is curative in the treatment of AP, macrophages, as an important component of the AP inflammatory cells, has not been thoroughly studied. This topic adapts the study of Gata6 dependent F4/80 + ICAM2+ and Gata6 independent F4/80 lo MCHII+ macrophages as the main line in the AP mice model. With application of various techniques and methods, such as immunofluorescence staining, FCM, the injection of clodronateliposomes, cell co-culture and protein chip and so on, it is to explore whether Gata6 dependent/independent macrophages regulate and how to adjust the AP injury recovery and the curative effect of traditional Chinese medicine Qingyi decoction, even to clarify which molecular mechanisms from different angles of animals, cellular and molecular expression. The study is to illustrate the recovery mechanism of AP injury from the perspective of the macrophage , and to explore the effective mechanism of TCM prevention by treatment of Qingyi decoction, it will be importantly clinically significant to understand the AP development and guide therapy, and make the motherland traditional Chinese medicine (TCM) to consolidate and the basic theory to develop, it provides new ideas and new clues for the prevention and control of major difficult diseases, and promotes the development of combination of Chinese and western medicine to a higher level.
急性胰腺炎(AP)是临床上常见的急腹症之一。清胰汤是临床疗效确切治疗AP的疗法,巨噬细胞作为AP炎症细胞的重要组分仍没有被研究透彻。本课题采用大鼠AP模型,以Gata6依赖性F4/80+ICAM2+和Gata6非依赖性F4/80loMHCII+巨噬细胞亚群研究为主线。应用免疫荧光染色、FCM、氯磷酸盐去除巨噬细胞、细胞共培养和蛋白芯片等多种技术和方法,从动物、细胞和分子表达不同水平探究Gata6依赖性/非依赖性巨噬细胞是否参与调控以及如何调控AP损伤修复和中药清胰汤疗效,并阐明其中分子机制。该研究从巨噬细胞亚群角度阐明AP损伤修复机制,探讨中医清胰汤防治AP的有效机制,对了解AP发生发展和指导治疗有重要的临床意义,并使祖国中医药学基础理论得到夯实和发扬,为防治重大疑难疾病提供新思路和新线索,推动中西医结合医学向更高层次发展。
急性胰腺炎(AP)是临床上常见的急腹症之一。清胰汤是临床疗效确切治疗AP的疗法,巨噬细胞作为AP炎症细胞的重要组分仍没有被研究透彻。本课题采用大鼠AP模型,以GATA6依赖性/非依赖性巨噬细胞亚群研究为主线。应用FCM、活细胞工作站、ELISA、RT-PCR和细胞共培养等多种技术和方法,从动物、细胞和分子表达不同水平首次探究AP损伤修复和中药清胰汤疗效中GATA6依赖性/非依赖性巨噬细胞动力学,表型和功能的变化,验证了GATA6依赖性巨噬细胞相比于GATA6非依赖性巨噬细胞更有利于AP损伤修复,阐明清胰汤通过调控两者细胞的动态平衡来达到治疗效果的机制,并发现关键分子TMEM195。该研究从巨噬细胞亚群角度阐明AP损伤修复机制,探讨中医清胰汤防治AP的有效机制,对了解AP发生发展和指导治疗有重要的临床意义,并使祖国中医药学基础理论得到夯实和发扬,为防治重大疑难疾病提供新思路和新线索,推动中西医结合医学向更高层次发展。
