The beet armyworm, Spodoptera exigua, is an economically important polyphagous insect pest all around the world. Metamorphosis is a complex, highly conserved and strinctly regulated development process that involves the programmed cell deth of obsolete larval organs. The developmental process of different larval tissues during metamorphic transformation showed that tissues such as the silk gland and gut are completely histolyzed by cathepsin, while other tissues such as fat body undergoes recoganization with histolysis. Cathepsin plays an important role in the developmental process and metamorphosis of insect. Here, we will focus on cathepsin D, a lysosomal aspartic proteinase in Spodoptera exigua, as a metamorphosis-specific proteinase involved in metamorphic events. To help elucidate the molecular mechanisms of metamorphosis in Spodoptera exigua, we cloned the Spodoptera exigua cathepsin D (SeCatD) gene from Spodoptera exigua. We will examine the expression profile of SeCatD during development process by RT-qPCR and Western Blotting. In order to determine which factors impact SeCatD expression, high temperature, ecdysone, juvenile hormone, insect pathogenic microorganism infection and viral infection on SeCatD mRNA expression will be examined. RNA interference technology will be used to identify the functional role of SeCatD in metamorphosis. The main goal of the project is to develop a novel potential pest control technology through insect physiological interference.
甜菜夜蛾(Spodoptera exigua)是一种世界范围内广泛分布、以危害蔬菜为主的杂食性、暴食性害虫。昆虫体内的组织蛋白酶参与昆虫体内各种生理生化反应和个体发育过程,其表达和分布部位大多位于脂肪体、幼虫消化道、早期胚胎及不同发育阶段的卵母细胞等,这些组织蛋白酶与昆虫的个体发育、组织分化以及变态反应密切相关。本研究以甜菜夜蛾cDNA文库中分离到的组织蛋白酶D基因为对象,通过实时荧光定量PCR技术、Western Blotting和RNA干扰技术对甜菜组织蛋白酶D的时空表达以及基因功能开展深入研究,阐明组织蛋白酶在甜菜夜蛾个体发育和组织分化过程中的功能和表达调控机制,为组织蛋白酶在昆虫变态反应中的研究提供参考,同时改变甜菜夜蛾体内组织蛋白酶的表达对其进行生理干扰,为有害生物防治技术提供新的思路。
本项目以蔬菜害虫甜菜夜蛾为研究对象,研究了其组织蛋白酶D及生长抑制多肽在甜菜夜蛾幼虫生长发育中的功能,取得如下成果:获得了甜菜夜蛾组织蛋白酶D基因全长序列,比对分析表明其在昆虫进化中高度保守;采用重组杆状病毒系统表达并纯化得到重组甜菜夜蛾组织蛋白酶D,揭示了其酶学特性;研究了甜菜夜蛾组织蛋白酶D在其生长发育中的表达规律,通过RNA干扰下调幼虫时期甜菜夜蛾组织蛋白酶D的表达,致使甜菜夜蛾幼虫变态异常。以上研究结果阐明了甜菜夜蛾组织蛋白酶D在甜菜夜蛾个体发育和组织分化过程中的功能,丰富了组织蛋白酶在昆虫变态反应中的作用机制。同时干扰甜菜夜蛾体内组织蛋白酶的表达能影响其生长与发育,为有害生物防治技术提供新的思路。此外,在研究的过程中发现甜菜夜蛾生长抑制多肽可影响其幼虫的生长发育。并首次从甜菜夜蛾体内克隆到甜菜夜蛾生长抑制多肽,阐明了其在甜菜夜蛾生长发育中的作用。研究发现,该基因表达受激素调控;重组甜菜夜蛾甜菜夜蛾生长抑制多肽能够显著抑制甜菜夜蛾幼虫的生长与发育;和其他同类型生长抑制多肽相比,重组甜菜夜蛾生长抑制多肽能够结合并抑制昆虫病原真菌白僵菌的生长,这为研究细胞因子抑制肽在免疫应答机制中的作用提供了新的视野。同时,重组甜菜夜蛾生长抑制多肽对甜菜夜蛾幼虫的生长具有良好的抑制活性并显著提高杆状病毒的杀虫活性,为生长抑制多肽在害虫生物防治中的应用奠定了基础。
