Telocyte is a novel type of interstitial cell that has been involved in the movement, blood supply, and immunity of various organs. In the present project, the entry point of the study is the characteristics of telocyte and their possible role. And the oviduct of laying hens during different postnatal periods was selected as research model. The aim is to explore the distribution and structural features of telocyte and the structural basis of targeting regulation between telocyte and their adjacent cell by cellular and molecular technology. At the same time, the difference of protein and mRNA expression about the markers of telocyte and adjacent cell will also be explored by RT-PCR and western blot in the oviduct of laying hens during different postnatal periods, which is aim to quantitative analysis of changing role of these cell during different postnatal periods. Meanwhile, in virto studies the potential function of telocyte will be analysed by live cell staining, Cell-IQ dynamic observation, iTRAQ, and then the result is compared and validated with their structural basis. The spatial structure and variation of the telocyte network in the oviduct of laying hens during different postnatal periods and their role of targeting regulation in neural regulation, blood flow control and immunoregulation will be examined at cellular, protein and molecular levels. This study provides a new idea for the cognition and treatment of diseases related to egg laying and reproduction disorders.
Telocyte是新近发现的一种间质细胞,与器官的运动、血供、免疫等重要活动密切相关。本项目拟以telocyte的特性为切入点,选择胚后发育不同时期的蛋鸡输卵管作为研究对象,运用细胞和分子生物学等技术,探究telocyte在蛋鸡输卵管的分布规律和结构特征,以及其靶向调节毗邻细胞的结构基础。并利用RT-PCR、Western blot技术,检测蛋鸡胚后发育不同时期输卵管telocyte及毗邻细胞标记物的蛋白和mRNA表达水平,定量分析这些细胞胚后发育的变化规律。同时进行离体实验,利用活细胞染色、Cell-IQ动态观察,iTRAQ手段分析telocyte的潜在功能,与结构基础进行比照和互为验证。在细胞、蛋白和分子水平上揭示胚后不同时期蛋鸡输卵管中telocyte网络的空间构筑和变化规律,及其在神经调控、血流控制和免疫调节中的靶向调节作用,为蛋鸡在产蛋和繁殖障碍类疾病的认知和治疗提供新的思路。
