Our country is facing a challenge of aging. According to the main data bulletin (no. 1) of the sixth national population census in 2010, aging speed of our people was the most quickly in the world. Diseases prevention and health care has become an important subject urgently to be solved by the field of medicine. As early as neijing era of traditional Chinese medicine "cure not ill" was proposed, and preventive acupuncture as the main means of "cure not ill", will play a great potential in the field of disease prevention..The study is guided by the theory of "cure not ill", and focuses on Alzheimer’s Disease (AD), which is the important problem in the aging society .At the same time, we will take advantage of "anti-aging gene" Klotho, which is new targets for diseases associated with aging in the world. After treatment, we will observe the influence of preventive acupuncture on cognitive ability of AD model mice. And then, we well use expression spectrum technology to test the Klotho gene and multiple genes related regulatory pathways for the efficient screening. for looking for the key ways of Klotho in prevention of AD, and also using Western blot technique to test Klotho and the key indicators of Klotho effect way for validation of Klotho in regulation function to prevent AD. In order to perfect the mechanism of action of dementia, for preventing AD provide scientific experimental basis for clinical application of preventive acupuncture, then to find new targets which will prevent age-related diseases .
据《2010年第六次全国人口普查主要数据公报(第1号)》显示,我国老龄化速度居世界之首。老年病的预防已成为目前医学领域亟待解决的重要课题。中医早在《内经》时代就提出“治未病”,而逆针灸作为其主要手段,将在预防领域发挥巨大潜力。.本课题以“治未病”为指导思想,针对老龄化社会的重要难题——老年性痴呆(AD),引入国际上治疗老化相关疾病的新靶点—“抗衰老基因”Klotho,观察逆针灸对模型小鼠认知能力的影响,同时采用基因表达谱技术对Klotho及其相关调控通路的多个基因进行高效筛选,寻找Klotho在预防AD过程中的关键途径,并利用Western blot 技术对Klotho及筛选出的Klotho作用途径中的关键指标进行检测,验证Klotho在逆针灸预防AD过程中的调控作用,完善其抗痴呆的作用机制,为临床应用逆针灸预防AD提供科学实验依据,进而为寻找逆针灸预防衰老相关疾病的新靶点提供线索。
老年性痴呆(AD)是老龄化社会的重要难题,本课题主要研究逆针灸保护认知能力、延缓AD发生的作用机制。课题选用6月龄雄性APPswe/PS1dE9 双转基因小鼠作为研究对象,围绕逆针灸对AD模型小鼠脑内抗衰老基因Klotho 的影响展开。综合行为学测试、基因表达谱筛选以及Western blot检测结果,逆针灸能够改善认知能力,上调Klotho水平,通过激活NF-κB 及Jak-STAT信号通路,在与认知功能维持相关的多个衰老过程中发挥重要的调控作用,进而有效防治AD。
