The reduction of endometrial receptivity and low embryo implantation rate which were result from ovulation induction were very difficult questions in assisted reproductive technology medicine. The role of Traditional Chinese Medicine in preventing miscarriage after embryo transplantation can not be ignored. But its mechanism has not yet clear. HOXA10 is a key gene of regulation during embryo implantation,and plays a leading role in the formation of endometrial receptivity. Related researches indicated that EG-VEGF、Ang-2 which were angiogenesis facto,might be involved in the formation of endometrial receptivity. But the further concrete mechanism is unknown. As our preliminary study showed,TCM could promote HOXA10 ,Ang-2 and EG-VEGF gene expression in primary culture of endometrial stromal and glandular cells. On this foundation,the study will establish an implantation animal model after ovulation induction and adhesion model in vitro,use the gene recombinant technology and RNA interference technology,study the interactions and the relationship between HOXA10 and Ang-2、EG-VEGF and their receptors,and discuss the control mechanism in angiogenesis of HOXA10 in endometrial receptivity. And expounds the mechanism of TCM in improving endometrial receptivity, may provide effective methods and reliable basis to improve the clinical pregnancy rate in assisted reproductive technology medicine.
促排卵治疗导致的子宫内膜容受性降低、胚胎低种植率是辅助生殖技术领域所面临的棘手问题。中药在胚胎移植术后的保胎作用不可小视,其作用机制尚未明确。同源框基因10(HOXA10)是调控胚胎着床的关键基因,在决定子宫内膜容受性方面起主导作用。研究表明,促血管生成素2(Ang-2) 、内分泌源性血管内皮生长因子(EG-VEGF)作为血管生成因子,可能参与调控子宫内膜容受性形成,但具体机制不明。本项目前期研究结果提示,中药可上调原代培养子宫内膜间质及腺上皮细胞HOXA10、Ang-2及EG-VEGF基因表达。本项目将在此基础上,建立促排卵后着床动物模型和体外粘附模型,采用基因重组技术、RNA干扰技术研究HOXA10与Ang-2 、EG-VEGF及相关受体间相互作用关系,探讨HOXA10在子宫内膜容受性形成过程中血管生成的调控机制,阐明中药干预的作用。为辅助生殖提高临床妊娠率提供有效的方法和可靠的依据。
