The contamination of mycotoxin in feed can result in serious problems on feed and food safety, therefore, it is extremely important to monitor mycotoxin level in feed for ensuring animal and human health. Surface enhanced Raman spectroscopy (SERS)-based nanobiosensor has the potential for multiplexing and rapid detection of mycotoxin in feed. However, how to simultaneously detect different mycotoxin at their respective regulation levels is one of the major obstacles for multiplexing detection. In this proposal, using fumonisin as a model chemical, we will firstly explore approaches for controllably adjusting hapten intensity on gold substrate, surface Raman label intensity and antibody intensity on SERS nanoprobe, then we will systematically study the effects of these factors on the apparent affinity constant between SERS nanoprobe and solid-phase probe, and further reveal the effects of these factors on assay sensitivity. Based on these results, we will finally clarify the mechanism for controllably adjusting assay sensitivity of SERS nanobiosensor. The fulfillment of this project will provide theoretical and practical support for the multiplexing detection of mycotoxin in feed, and moreover, it will also provide a novel idea and method for the optimization of assay sensitivity in other immunoassay development.
饲料中霉菌毒素的污染可引起严重的饲料和食品安全问题,因此监控其污染水平对保障动物和人类健康具有重要意义。基于表面增强拉曼光谱(SERS)的纳米生物传感器具备同步快速检测多种霉菌毒素的能力,然而如何实现各组分灵敏度的可控调节以同时满足不同组分在限量水平的检测要求,是霉菌毒素同步分析亟待解决的关键问题之一。在前期已初步构建SERS纳米生物传感器分析平台的基础上,本申请拟以伏马毒素作为模式化合物,探讨可控调节固相探针表面半抗原密度、SERS纳米探针表面拉曼标记物和表面抗体密度的方法,系统研究各参数对固相探针与纳米探针相互作用表观亲和常数的影响,并进一步揭示其对SERS纳米生物传感器分析灵敏度的影响规律,从而阐明可控调节该平台分析灵敏度的机制。本申请创新性地提出灵敏度可控调节的概念,研究结果不仅可为饲料中霉菌毒素的同步分析提供理论和技术支撑,并且可为其它免疫分析灵敏度的优化研究提供新的思路和方法。
饲料中霉菌毒素的污染可引起严重的饲料和食品安全问题,因此监控其污染水平对保障动物和人类健康具有重要意义.针对饲料中不同霉菌毒素共污染和污染水平差异大的特点,本研究开发了可同步检测多种霉菌毒素的多组分纳米金免疫层析试纸条方法,以及基于表面增强拉曼光谱的免疫传感分析方法,同时系统研究了固相探针表面半抗原密度、纳米探针粒径和表面抗体密度等对免疫分析灵敏度的影响规律,提出了多组分免疫分析灵敏度可控调节的思路,实现了对不同霉菌毒素在相应限量水平的同步快速检测。该方法可显著降低检测成本,提高检测效率,同时灵敏度可控调节的机制研究丰富了免疫分析技术的理论和实践。研究结果在Food Chemistry(IF4.529)杂志发表论文3篇,申请发明专利1项。
