中药经肠道干预慢性肾功能衰竭大鼠模型的肠道上皮细胞紧密连接的研究

Chronic kidney disease is a public health problem with worldwide concern, about 5% of patients with chronic kidney disease who will eventually round into the chronic renal failure. But in the progression of chronic renal failure, lacking of effective interventions. Contemporary Chinese medicine have made many useful attempts to slow the progress of chronic renal failure by application of traditional Chinese medicine by intestinal. Studies have shown that in addition to reducing ure, creatinine, middle molecular substances toxins such as level, the treatment by intestinal still delay the progress of chronic renal failure . But its mechanism of action and therapeutic targets is not clear. The subject using Chinese medicine treatment by the intestine on chronic renal failure rat model, with other methods of Realtime PCR,Western blot laser scanning, renal biopsy and so on, lucubrating the mechanisms and therapeutic targets of Chinese medicine treatment by intestinal . Observeing the change of the intestinal tight junction protein and renal pathology, to explore new theoretical basis and treatment direction of Chinese medicine to chronic renal failure.

慢性肾脏病是全世界关注的公共卫生问题之一，有约5%的慢性肾脏病患者最终将进入慢性肾功能衰竭（CRF），但在慢性肾脏病的进展过程中，缺乏有效的干预措施。当代中医在临床上应用中药经肠道治疗CRF，在延缓CRF的进展方面做出了许多有益的尝试, 研究显示除了降低CRF患者的尿素、 肌酐、 中分子物质等毒素水平，还在延缓该病进展方面显示出一定疗效，但是其作用机制和治疗靶点尚不明确。本课题采用CRF大鼠模型行中药经肠道治疗，使用Realtime PCR、Western blot、激光扫描及肾脏病理检查等方法，通过观察治疗前后肠道上皮细胞紧密连接蛋白的结构和功能的变化、肾脏病理的前后变化，深入研究中药通过干预肠道上皮细胞紧密连接来延缓CRF进展可能的作用机制及治疗靶点。为中药经肠道治疗延缓CRF的进展探寻新的理论基础和治疗方向。
