stress induced-senescence cells may induce and accelerate the aging of the body and organization, while autophagy can clear damaged structure in the cells . Therefore, the relationship between autophagy and aging has been a new hot area of research now.HSF is the major cell types in the dermis, aging HSF cells not only is the product of skin aging, also may be the reason for the further skin aging. The study of the relation between autophagy and HSF aging is scanty. It is necessary to find targeted repair methods of aging HSF by regulating autophagy. This topic will study the effect and mechanism of Yiwei Gubiao therapy in regulation of autophagy of stress induced-senescence of HSF by the PI3K/Art/mTOR signaling pathway. Discusses how to use traditional Chinese medicine regulating autophagy Appropriately. This study can provides experiences for delay skin aging and age-related disease. providing theoretical and experimental basis for the application of yupinfeng powder and development of new drugs of Chinese medicine in prevention and treatment of skin-aging.
衰老细胞的积聚会诱发并加速机体和组织的衰老,而细胞自噬则能通过清除细胞中的受损结构,减缓或修复细胞衰老。因此,自噬与衰老相关性的研究,已成为医学抗衰领域中的新热点。HSF是皮肤真皮中最主要的细胞类型,衰老HSF不仅是皮肤衰老的产物,也可能是皮肤进一步衰老的原因。但目前针对自噬与HSF衰老相关性的研究不多,缺乏中医药对衰老细胞自噬调控的研究。所以有必要寻求良好的具有针对性调控细胞自噬从而修复或清除衰老HSF的方法及药物。本课题拟采用血清药理学方法,以应激性衰老HSF为研究对象,以细胞自噬相关指标为研究内容,基于PI3K/Akt/mTOR信号通路研究益卫固表法对应激性衰老HSF自噬活性的调控效应及机制,并进一步探讨如何通过中医药诱导适度的自噬活动从而延缓衰老进程,为延缓皮肤衰老及衰老相关疾病中医药研究积累可以借鉴的研究思路与方法。预期成果将有益于皮肤衰老的防治和中医经典方剂玉屏风散的开发。
