整合素αvβ6抗体对角膜基质纤维化的作用及机制研究

Corneal matrix fibrosis can cause irreversible visual impairment and seriously threaten the visual acuity of patients. There is still a lack of effective anti-corneal fibrosis drugs. In our previous study, the integrin αvβ6 gene was knocked out in mouse and the corneal stromal fibrosis was obviously inhibited. However, whether the integrin αvβ6 antibody can effectively inhibit the fibrosis of human corneal stromal cells is still unknown. Therefore, the project will use the integrin αvβ6 antibody to observe whether it has adverse effects on the morphology of corneal stromal cells, whether it can effectively inhibit TGF-β1, inhibit the synthesis or secrete of α-SMA and collagen III, thus inhibiting corneal fibrosis, and whether the αvβ6 antibody has adverse effect on the collagen I and keratocan of normal healing secretion. The purpose of this study is to observe whether the integrin αvβ6 antibody can effectively inhibit the fibrosis of human corneal stromal cells without adverse effects and its mechanism, it may provide a new idea for the treatment of corneal matrix fibrosis.

角膜基质纤维化可造成不可逆的视功能损害，严重威胁患者视力。目前尚缺少有效抗角膜纤维化药物。本课题前期研究中敲除小鼠整合素αvβ6基因，可观察到角膜基质纤维化明显受到抑制。但整合素αvβ6抗体是否能有效抑制人角膜基质细胞纤维化还是未知。因此本课题将使用整合素αvβ6抗体，观察其对人角膜基质细胞形态有无不良影响；是否可以有效抑制TGF-β1，抑制角膜基质细胞合成或分泌α-SMA，collagen III，从而抑制角膜纤维化；以及整合素αvβ6抗体对正常愈合分泌的collagen I，keratocan有无不良影响。本研究的目的是观察整合素αvβ6抗体能否有效抑制人角膜基质细胞纤维化及其作用机制，以及其对角膜基质细胞有无不良影响，为角膜基质纤维化的治疗提供新思路。

课题组对研究发现整合素抗体对角膜基质纤维化有明显的影响作用。通过抑制TGF-b1的激活从而抑制角膜基质a-SMA的产生，可以改善胶原纤维III的含量，延缓角膜基质纤维化的形成过程。同时课题组对SMILE手术过程中取出的近视角膜基质进行了蛋白组学研究，在国际上及国内首次发现在中度近视和高度近视人群角膜角蛋白2和角蛋白10含量明显低于低度近视人群，目前国内外无相关报道。同时课题组发现高度近视人群的铁代谢异常，研究结果也为国际创新。研究结果申请国家发明专利5项，已经授权1项，4项处于实质审查阶段。发表SCI论文2篇，还有一篇正在审稿中。这些研究结果对于中度近视高度近视人群的药物治疗有重要的临床研发价值。靶向调控角膜角蛋白2和角蛋白10的含量对于近视的精准治疗或许有重要的研发价值，也为近视机制的研究提供了新的思路。
