Chronic pain has become a great threat to people health.Research has proved that DNA hypermethylation exists in pain states .There is a close relation between chronic pain and the hypermethylation of the promoters of genes which is relative with GABAergic neurons .Our early research work shows the mechanism of acupuncture analgesia is relative with improving the neuron plasticity .The aim of the research is to discuss the influnce of electro-acupuncture "Zusanli"and"Yanglingquan" to the function of GABAergic neurons ,the level of global DNA methylation,and DNA methylation of GABAergic neurons in spinal doral horn of rat model of sciatic nerve chronic constriction injure.The methods of patch clamp is used to detect to the function of GABAergic neurons ,Elisa is used to detect the level of the global DNA methylation,real time PCR and Western blot are used to detect the expression of Dnmt ,MeCP2, GABA(A)、(B)receptors、GAD67 and the corresponding expression modified by DNA methylation.The research result would play a import role in elucidating the mechanism of electroacupuncture analgesia.
慢性疼痛严重威胁人类健康。研究表明,DNA高甲基化在慢性疼痛中广泛存在,其中GABA能神经元相关基因启动子高甲基化与慢性疼痛关系密切。前期工作基础表明,针刺足三里、阳陵泉治疗坐骨神经痛机制与改善神经元可塑性有关。本课题主要研究电针"足三里"、"阳陵泉"对慢性坐骨神经痛模型脊髓背角GABA能神经元功能、DNA甲基化水平以及GABA能神经元相关DNA甲基化 的影响,我们采用膜片夹实验检测GABA能神经元微电流水平,免疫组化检测总DNA甲基化水平、实时定量PCR法和Western blot法检测Dnmt在GABA能GABA(A)、(B)受体、GAD67的表达及其启动子甲基化水平,荧光免疫组织化学双重染色法检测Dnmt1、Dnmt3a、 Dnmt3b与GAD67在GABA能神经元的表达。研究结果对阐明电针镇痛的机制具有重要意义。
慢性疼痛严重威胁人类健康。研究表明,DNA高甲基化在慢性疼痛中广泛存在,其中GABA能神经元相关基因启动子高甲基化与慢性疼痛关系密切。本课题主要研究电针”ST36”-“GB34”对慢性坐骨神经痛模型脊髓背角GABA能神经元功能、DNA甲基化水平以及GABA能神经元相关DNA甲基化的影响。研究结果显示:1.电针“ST36”-“GB34”可以减轻慢性疼痛,电针镇痛效果优于Dnmt抑制剂5-AZA。2.电针“ST36”-“GB34”可以上调CCI引起的GABA(A)、(B)受体、GAD67mRNA的表达及血清GABA含量下降;改善CCI引起的大鼠脊髓背角GABAA Rβ1、GABAB、GABAB R1、GAD67阳性表达下降,从而调整CCI模型大鼠脊髓背角GABA能神经元功能。3.电针“ST36”-“GB34”可以降低CCI后引起的Dnmt1、Dnmt3a、 Dnmt3b、MeCP2 mRNA表达上调及CCI后引起GABA(A)、(B)受体、GAD67启动子区CpG岛的甲基化水平和脊髓腰膨大处总体甲基化上升,并且电针组优于Dnmt抑制剂5-AZA组。电针可以降低CCI后引起大鼠脊髓背角Dnmt1、Dnmt3a、MeCP2与GAD67在GABA能神经元的表达上调。科学意义:电针ST36、GB34通过降低脊髓背角总体甲基化水平,抑制脊髓背角GABA能神经元相关DNA甲基化,提高脊髓背角GABA能神经元功能,达到镇痛的作用。对阐明电针镇痛的机制具有重要意义。
