Erectile dysfunction (ED) is a major complication of diabetes, and many diabetic men with ED are refractory to common ED therapies. The lack of NGF is an important factor of diabetes ED(DMED). Stem cells hold great promise for regenerative medicine because of their ability to self-renew and to differentiate into various cell types. Meanwhile, in preclinical experiments, therapeutic gene-modified stem cells have been approved to offer a novel strategy for cell therapy and gene therapy of erectile dysfunction. The human umbilical cord mesenchymal stem cells (HUMSCs) have been reported to secrete various cytokines that exhibit angiogenic and neurosupportive effects. and is a good choice for gene therapy carrier. This study was conducted to investigate the effects of HUMSCs modified with nerve growth factor on DMED in rats.
糖尿病勃起功能障碍(DMED)是糖尿病引起的泌尿系统常见并发症,因临床治疗缺乏针对性,只能暂时缓解症状。NGF减少是DMED重要的致病因素。我们前期实验中发现糖尿病大鼠阴茎组织中NGF表达较正常大鼠明显减少,我们前期试验中使用的脐带间充质干细胞(MSCs)既具有多向分化潜能,又是一种良好的基因治疗的载体细胞。在此基础上课题组拟在构建含人NGF的重组慢病毒载体,采用基因转染技术构建含有重组慢病毒载体的人脐带间充质干细胞,使人脐带间充质干细胞持续稳定的表达NGF,从而使转NGF后的人脐带MSCs分别发挥基因治疗和细胞治疗作用。利用DMED动物模型检测转NGF基因人脐带间充质干细胞作用前后阴茎海绵体相关因子及基因的变化,明确其对DMED的作用机制,为DMED的治疗开辟新途径。
