More than 100 new sesquiterpenoids, with 4 novel skeletons, were isolatrd and identified from 19 medicienal plans of Compositae icluding a new genus with single species of Ligulariopsis from the particular plant in northwest China, the new genus close related with Ligularia genus, Calcalia genus and Cnemanthodium genus, etc.The antitumor activity and the inhibition of telomerase activity of sesquiterpenoids in 3 tumor cell lines were assayed by traditional MTT method and ELISA method respectively. Among them, some compounds exhibited potertial antitumor activity, however, does not every one exhibit effect on telomerase activity simultaneously.Thefore , the antitumor activity can not evaluated solely by inhibitory effect on telomerase activity, While understanding the regulation of telomerase may still provide insights for developing novel cancer therapeutic approach. The property of this study is that the natural product chemistry is closely cooperated with systemalic botant and pharmacolgy.The results showed an important scientific basis for developing and utilizing the natural resoures and searching new antitumor diug.
以端粒酶新靶点寻找抗癌、抗癌活性的天然萜类和先导物,这是一种简洁、快速、专一、灵敏的新方法。本课题用端粒酶跟踪筛选与化学研究相配合,运用现代分离、分析技术对几种垂头菊、橐吾属植物中倍半萜等活性成分进行分离、结构鉴定、药理和构效关系等研究,是一项创新性课题,对发现新型抗癌、抗衰老活性物质及开发新药,具有重要战略意义。
