Spinal cord glioma is the most common intramedullary tumor. Tumor resection always leads to severe neurological damage. The postoperative radiotherapy and chemotherapy are not ideal with high recurrence rate. Therefore we proposed the time controlled release drug stent with different drug coating package stent fiber, early delivery of chemotherapy drugs to kill the residual tumor cells and prevent recurrence, late delivery of neurotrophic factors promote the recovery of nervous system, and the stent fibers may also play a role in nerve repair guiding. This research is mainly to study the making method and biological safety of time controlled release drug stent in the spinal cord glioma treatment, including of technical process and parameters of PLGA/ collagen nano stent, by the layer-by-layer self-assembly technique for nano coating containing temozolomide and NT3 growth factor on fiber surface, the efficacy and safety in treatment of the rat spinal cord glioma animal model.The time controlled release drug stent provides a new therapeutic approach for the treatment of spinal cord glioma and other intramedullery lesions through packaging different drug, which will improve the treatment of spinal cord disease, has the vital significance to reduce the spinal nerve dysfunction.
脊髓胶质瘤是最常见的髓内肿瘤,手术切除对神经传导系统破坏较大,常留有严重的神经功能障碍,术后复发率高,目前术后的放化疗效均不理想。因此我们提出了分时控释脊髓支架的思路,在纳米支架纤维表面封装不同的药物涂层,早期缓释化疗药物杀灭残余肿瘤细胞防止复发,后期缓释神经营养因子促进神经系统的修复,而支架本身对正常神经纤维的修复也可起到导向的作用。本课题拟对脊髓纳米载药分时控释支架的表征效应参数及在脊髓胶质瘤综合治疗中的生物机制进行动物实验研究。探索PLGA/胶原纳米多孔脊髓支架的技术流程和相关参数性能,通过层层自组装技术在支架纤维表面构建包含替莫唑胺和NT3生长因子的纳米涂层,在大鼠脊髓胶质瘤动物模型上验证其疗效和生物安全性。脊髓纳米载药分时控释支架将为脊髓胶质瘤的治疗提供新的治疗途径,通过封装不同的药物,可以应用到其它髓内病变的治疗中,这将为提高脊髓疾病疗效,降低脊髓神经功能障碍具有重要的意义。
