Lianhua qingwen capsules is a Chinese traditional medicine researched by the guidance of collaterals disease theory, which has the function of broad-spectrum antivirus. The animal experiment and clinical research indicated that Lianhua qingwen capsules was quite effective on influenza, however the efficacious material basis hasn’t been revealed. In this project, we will ascertain the main constituents by UPLC-MS and establish the chemicalome and ingredient library. Based on serum pharmacochemistry, we will analyze the rat serum obtained after oral administration of Lianhua qingwen capsules to ascertain the Lianhua qingwen capsules-related components, the activities of the Lianhua qingwen capsules-related components in the serum will be screened using in vitro anti-influenza virus pharmacological models to elucidate the material basis for efficacy and mechanism. Through this subject, we could not only provide a base on instituting a more scientific quarlity standerd of Lianhua qingwen capsules and its secondary development, but also help on the mordenization of traditional Chinese medicine and the research of material basis for efficacy of Chinese herbal formulas.
连花清瘟胶囊是运用络病理论空间概念探讨外感温病及瘟疫病机传变规律,提出积极干预治疗对策研制的组方,具有广谱抗病毒作用,动物实验和临床结果均表明对流行性感冒具有良好的治疗效果,然而其抗流感病毒的药效物质基础尚不明确,本研究拟采用UPLC-MS对连花清瘟胶囊中主要化学成分进行全面系统的表征,构建连花清瘟胶囊化学成分谱和化学成分库,利用“血清药物化学理论”结合UPLC- MS技术对动物口服连花清瘟胶囊后入血成分进行定性分析,确定体内药源性成分,并对体内药源性成分进行体外抗流感病毒评价,从而确切阐明连花清瘟胶囊的药效物质基础及作用机理。本研究不仅可为连花清瘟胶囊科学化的质量标准的制定、二次研究开发提供有力的科学依据,同时宜可为推进中药现代化,中药复方药效物质以及作用机理研究提供有效的参考。
本项目对连花清瘟胶囊的药效物质基础进行了系统的研究,通过化学成分研究,从连花清瘟胶囊中分离并鉴定了87个化合物,构建了连花清瘟化学成分库,利用UPLC/Q-TOF-MS/MS技术对动物口服连花清瘟胶囊后入血成分进行定性分析鉴定了22个成分,对22个入血成分和分离得到的单体化合物进行了抗流感病毒活性测试,7个化学成分能部分抑制H3N2流感病毒CPE的产生,2个化学成分部分抑制H1N1流感病毒CPE的产生,且进行了流感病毒M基因拷贝数定量检测,初步阐明连花清瘟胶囊抗流感病毒的物质基础和作用机理。较为圆满地完成了计划任务。
