Many of inflammatory cells infiltrated in tumor microenvironment, among them mast cells have both tumor promoting and anti-tumor roles. In the previous study, we have demonstrated that bone marrow derived mast cells (BMMCs) could accelerate growth of 4T1 breast cancer, however through IgE receptor FcεRI activated BMMCs reversed their pro-tumorigenic role. Preliminary mechanism study suggested that activated BMMCs inhibit 4T1 cell proliferation and migration, block PI3K/Akt signal pathway. . In this project using the molecular and cellular pharmacology and animal experiment methods to study anti-tumor activity of through IgE receptor FcεRI activated mast cells which infiltrated in tumor; clarify anti-tumor mechanism from BMMCs mediator by in vitro and in vivo, expect to find anti-tumor BMMCs mediator, provide experiment data and new strategy for therapy of cancer.
肿瘤的微环境中浸润多种炎症细胞,其中肥大细胞对多种肿瘤既有促进作用又有抑制作用。本课题前期发现骨髓来源的肥大细胞(BMMCs)对4T1乳腺癌具有促进作用,但其IgE受体FcεRI激活后对4T1肿瘤的生长具有抑制作用。初步机制研究表明激活的BMMCs可抑制4T1细胞的增殖、迁移及阻断PI3K/Akt信号通路等作用。. 本课题将采用分子细胞药理学方法和整体动物实验、深入全面地研究浸润到肿瘤中的肥大细胞通过FcεRI激活后所介导的抗肿瘤作用;通过多种体内外实验阐明所激活的BMMCs细胞因子抗肿瘤作用机制,以期发现具有抗肿瘤作用的肥大细胞因子,从而对临床治疗肿瘤提供实验依据及新的思路。
肿瘤的微环境中浸润多种炎症细胞,其中肥大细胞对多种肿瘤既有促进作用又有抑制作用。本课题发现骨髓来源的肥大细胞(bone marrow derived mast cells , BMMCs)对4T1乳腺癌具有促进作用,但其免疫球蛋白E(Immunoglobulin E, IgE)受体FcεRI激活后对4T1肿瘤的生长及迁移具有抑制作用。利用ELISA试剂盒及中和抗体发现激活的肥大细胞的抗肿瘤作用是通过肿瘤坏死因子(tumor necrosis factor α, TNFα)而产生作用的。小鼠体内动物实验进一步验证了通过FcεRI激活的肥大细胞介导的抗肿瘤作用,肿瘤组织的免疫组化及Western blot实验结果表明激活的肥大细胞有效抑制磷脂酰肌醇3-激酶(phosphoinositide 3 kinase, PI3K)/Akt通路的激活、分裂原活化蛋白激酶(mitogen activated protein kinases, MAPKs)的激活、肿瘤的血管生成及信号转导和转录激活因子3(signal transducer and activator of transcription 3, Stat3)的磷酸化激活等。以上结果为临床治疗肿瘤提供实验依据及新的思路,有望为肿瘤的免疫治疗奠定理论基础。
