Rose is a very famous traditional flower in China. Because of its rich flower type and color, many people like it. Rose is one of the most important cut flowers in the world flower market now. With the industrial technology innovation and the improvement of living standards, rose breeding for high quality has been paid more and more attention. Rose has complex genetic background. The genetic basis of ornamental traits are not clear now. High efficient MAS technique depends on high density genetic map and accurate positioning important traits. Therefore, rose high density genetic map construction and characters of accurate QTL positioning are very important. In this project, the genetic map were constructed based on a F1 population crossed from R. Wichuraiana and YJ-2007-16 using “pseudo-test cross” mapping strategy based on SSAP and RAD-Seq technology. The SSAP primers were designed by our laboratory. QTLs controlling receptacle size, flower diameter size will be detected. This project will provide scientific theoretical foundation on rose MAS breeding, gene cloning and genome research.
月季为中国传统名花,因其花色丰富,色彩艳丽,深受人们喜爱,为世界四大切花。随着产业技术创新和生活水平的提高,月季品质育种日益受到重视,但月季的遗传背景较复杂,目前对其重要性状的遗传基础了解还较少。高效的分子标记辅助育种技术依赖于饱和的遗传图谱和重要性状的准确定位。因此,月季饱和遗传图谱的构建和性状准确的QTL定位十分重要。本项目基于“拟测交”作图策略,以光叶蔷微(R. Wichuraiana)和YJ-2007-16为亲本,杂交所得F1群体为试材,利用本实验室所设计月季SSAP引物和RAD-Seq技术,构建月季高密度遗传图谱,并对花托大小、花径大小等性状进行QTL定位。该项目的进行将为月季分子标记辅助育种及今后月季图位基因克隆、基因组学等研究奠定基础。
月季为中国传统名花,因其花色丰富,色彩艳丽,深受人们喜爱,为世界四大切花。随着产业技术创新和生活水平的提高,月季品质育种日益受到重视,但月季的遗传背景较复杂,目前对其重要性状的遗传基础了解还较少。高效的分子标记辅助育种技术依赖于饱和的遗传图谱和重要性状的准确定位。本研究基于“拟测交”作图策略,以亲本杂交所得151个杂交个体为作图群体,利用SSAP和简化基因组测序,构建月季高密度遗传图谱。父本遗传图谱包括1470个标记,总图距为2961.75cM,标记间的平均距离为2.02cM。母本遗传图谱包括3931个标记,总图距为5918.54cM,标记间的平均距离为1.51cM。利用同源位点进行双亲图谱整合,最终得到的整合图谱共7个连锁群,与月季染色体数一致,包括5305个标记,总图距为4913.63cM,标记间的平均距离为0.93cM。该结果为相关性状的QTL分析提供了理论基础。
