The occurrence of chestnut blight severely affects the economic value of chestnut planting and the health of forest ecosystem. Castanea henryi and C. mollissima planting in China are important genetic resources of the genus Castanea to breed resistant trees, so it has important theoretical and practical significance to study comparative genomics between the two species. In this project, a high density genetic linkage map of C. henryi will be constructed using SNPs developed from resequencing and SSRs from the genus Castanea. Collinearity of blight resistance QTLs will be detected by comparative mapping between C. henryi and C. mollissima. The candidate genes and common SNPs and InDels will be identified by comparing the sequence of genome or blight resistance QTL regions of C. henryi and C. mollissima. This study will establish the relationship of genome information and reveal the similarities and differences of blight resistance loci between C. henryi and C. mollissima. The candidate genes and common markers of blight resistance will be helpful for molecular breeding of Castanea.
栗疫病的发生严重影响栗树种植的经济价值和森林生态系统的健康。我国栽培的锥栗和板栗是栗属树种抗病育种的重要基因资源,进行锥栗和板栗的比较基因组学研究,对于栗属树种的抗病育种具有重要的理论和实践意义。本项目将利用重测序开发的SNP标记和栗属SSR标记构建锥栗高密度遗传连锁图谱,通过比较作图探讨锥栗和板栗疫病抗性QTL的共线性,通过锥栗和板栗基因组及栗疫病抗性QTL区域序列的比较发掘与栗疫病抗性相关的候选基因及通用SNP和InDel标记。研究结果将建立锥栗和板栗基因组信息之间的联系,揭示锥栗和板栗疫病抗性位点的相似性和差异性,发掘的与栗疫病抗性相关的候选基因及通用标记为栗属树种分子育种提供参考。
