通过体外发酵,采用PCR/DGGE这一DNA指纹技术研究在不同日粮条件下瘤胃真菌和乳酸产生菌群之间的变化关系,分析乳酸浓度的消长与淀粉酶、纤维素酶、半纤维素酶活性的关系;同时通过动物试验,比较正常日粮和高淀粉日粮条件下瘤胃微生物区系变化以及L型和D型乳酸浓度的消长,分析瘤胃生理生化环境的变化,并通过瘤胃导入厌氧真菌试验,研究瘤胃真菌菌群对维持乳酸浓度的作用,揭示引起乳酸产生菌大量繁殖、乳酸大量积累的日粮因素和微生物区系不平衡因素,为解决瘤胃慢性酸中毒问题开辟一条新思路,同时为深入研究瘤胃调控、提高生产性能提供理论依据。
