The accomplishment of human genome project (HGP) will make the analysis of gene function becomes the core of research in post-genome era. The Drosophila becomes an excellent model organism for genetics, analysis gene function(s) and many biotechnologies arise. We constructed Gal4-UAS system, which is a good system to express exogenous gene and analyze its function. Firstly, we constructed the expression vectors (RP2-pUAST, RP2(del6)-pUAST, SNF1A-pUAST). Secondly, we transgened these plasmids into fly by microinjection. Finally, we balanced the transgenic flies and crossed the transgenic flies with Gal4 lines. .S2 cell transfection, PCR, Western blot and Fluorescence analysis detected the expression of these transgenes. These results show that human gene can express at fly. And we make sure the system of fly can analysis human gene's function by large scale.
