Based on the researches home and abroad, ATP and its metabolite adenosine play major roles in the primary regulation of acupuncture effects. As a kind of physical stimulation, acupuncture leads to the responsive changes of ATP and adenosine around the acupuncture points. But the detection technique for them nowadays has the disadvantage of tedious operation and time delay, which make it difficult to explain the effect of them in the acupuncture mechanism clearly. Based on the research platform of adjuvant-induced arthritis, this project plans to research the responsive changes of ATP and adenosine around the acupuncture points as well as the time-effect and dose-effect relationship of them by real-time monitoring biosensing technology; With the help of the neural electrophysiological method, their neurobiology mechanism of action in the primary regulation of acupuncture effects will be expounded. And we try to collect the data and establish mathematical model, in order to clarify the correlation between the responsive changes of ATP and adenosine, electrical signal feature of afferent nerve, and the effect of target organs. As a result, this project will be possible to supply a new modern biology method in researching the regulatory mechanism of acupuncture and provide the scientific basis for clinical translation.
国内外研究表明,ATP及其代谢产物腺苷在针刺效应初始调控中发挥着重要作用。作为一种机械性刺激,针刺可引起穴位局部ATP和腺苷的应答变化,但目前检测穴位局部ATP和腺苷的技术手段多存在操作繁琐、结果延时等局限性,尚不能清楚阐明二者在针刺起效中的作用。本课题拟采用能够实时、在体、连续检测的生物传感针技术,以针刺足三里治疗佐剂性关节炎大鼠模型为针刺效应平台,动态研究穴位局部ATP和腺苷的应答变化及其与针刺效应之间的时效、量效关系;结合神经电生理学方法,阐明二者在针效初始调控中的神经生物学作用机制;并进行数据挖掘,建立相关数学模型,明确“穴位局部ATP和腺苷——传入神经电信号特征——靶器官效应”三者之间的相关性。本课题的开展旨在为针刺效应原理研究提供更为直观有效的技术方法,探索针效调控的新模式,揭示针刺起效的部分作用规律,并进一步为促进临床转化、提高临床疗效提供科学依据。
