It has been widely indicated that MUC-1(mucin glycoprotein )and GlyCAM-1(glycosylated celladhesion molecule1)secreted from the epithelial cells of bovine, ovine and the other ruminants' uterus, can keep the normal uterus receptivity and implantation of their fertilized eggs on uterine endometrium. However,its basic function about the regulation of uterus receptivity on early pregnant of those sheep be artificially induced estrus, is still kept unknown. Therefore, in this study, both spatiotemporal transcription and expression of these two genes on sheep early pregnant utrine among the natural mating ( Control group ), only natural estrus, artificial induced estrus and mating, and only artificial induced estrus ones combining with detecting of their progesterone level in serum , will be further compared with one another, and analyzed systematically by In situ Hybridization, semi-quantative RT-PCR, Immunohistochemical Staining, Western Blot Hybridization, and Radioimmunoassay. These will be also helpful to ascertain the relation between the abnormal expressions of MUC1 and Glycam1 on ovine uterus receptivity for their early implantation of IVF embryos and pregnant failure.
家畜子宫腔上皮主要分泌的MUC-1(mucin glycoprotein,一种大分子转膜黏蛋白-糖蛋白)与 GlyCAM-1(glycosylated cell adhesion molecule1,糖基化细胞黏着分子-1)是调控正常子宫容受态、保障胚胎正常着床的先决条件。目前尚未见到其影响经人工诱导发情处理后的绵羊相应子宫容受态变化的研究报道;因此,本研究拟采用半定量RT-PCR,免疫组化,Western和放射免疫等方法,以本交配种母羊早期胚胎着床子宫中孕酮(P4)水平,以及MUC-1与GlyCAM-1基因表达变化为对照,对自然发情、人工诱导发情本交以及仅人工诱导发情处理的绵羊早期妊娠子宫中P4 水平,以及这两种基因在转录和翻译水平上,分别进行定性、定量表达比较,为进一步研究胚胎移植过程中,维持绵羊子宫正常容受态的相关基因可能出现的非正常表达与着床失败等生殖疾病的关系奠定基础。
研究表明,牛、羊等家畜子宫腔上皮主要分泌的MUC-1(mucin glycoprotein,一种大分子转膜黏蛋白-糖蛋白)与GlyCAM-1(glycosylated cell adhesion molecule1,糖基化细胞黏着分子-1)是调控其正常子宫容受态、保障体内胚胎正常着床的先决条件。但尚未见到有关这两种蛋白分子影响经人工诱导发情处理后的绵羊相应子宫容受态变化的研究报道;因此,本研究拟采用原位杂交、半定量 RT-PCR,免疫组化和Western blot等方法,以本交配种母羊早期着床部位子宫中 MUC-1与GlyCAM-1基因表达变化为对照,对自然发情、人工诱导发情并配种以及仅人工诱导发情处理的相同发情期绵羊子宫中这两种基因在转录和翻译水平上,分别进行定性、定量表达比较,我们的研究结果显示:在人工诱导发情绵羊子宫的第10 d到18 d,MUC-1和GlyCAM-1表达呈现一种先上升后降低的趋势,在第14 d达到最高表达量。对于MUC-1和GlyCAM-1的定位,在第10 d,12 d和14 d主要表达于子宫腔上皮上,第16 d和18 d主要表达于腺上皮。然而,在人工诱导发情后配种绵羊MUC-1和GlyCAM-1在第10 d和12 d表达量升高,在第14 d表达量却显著降低,之后再第16 d和18 d表达量再次升高。此外,MUC-1和GlyCAM-1在腔上皮和腺上皮上均有分布。在整体水平上,MUC-1和GlyCAM-1在人工诱导发情并配种母羊子宫内的表达显著低于在未配种母羊内的表达。除此之外,MUC-1和GlyCAM-1在自然发情与自然发情配种所得到的表达规律与人工诱导发情结果一致。我们的研究结果为进一步研究胚胎移植过程中,维持绵羊子宫正常容受态的相关基因可能出现的非正常表达与着床失败等生殖疾病的关系奠定基础。
