选择与牙周炎关系最为密切的四种可疑牙周致病菌:牙龈卟啉菌、放线共生放线杆菌,中间型普及菌和具核梭杆菌作为研究对象,提取染色体DNA,随机克隆入载体PUC19,转化大肠杆菌JM109,从中筛选出能与同种细菌染色体DNA进行特异性杂交而不与其它相近的细菌染色体DNA交叉杂交的克隆,作为该种细菌特异性的DNA探针。本课题筛选出PZJ-AaI,含2.1kb放线共生放线杆菌染色体DNA片段,PZJ-PgI,含1.8kb牙龈卟啉菌染色体DNA片段,PZJ-PiI,含2.7kb中间型普反菌染色体DNA片段,PZJ-FnI,含1.3kb具核梭杆菌染色体DNA片段,该四个克隆各与同种细菌染色体DNA呈特异性杂交反应,其敏感性在地高率标记下为10ng染色DNA,可以作为这中种可疑牙周病原菌的DNA探针。
