Continuous and extensive use of insecticides has led to the emergence and spread of insecticide resistance in insect populations, and thus deteriorated the spread of insect-borne disease. Understanding the molecular mechanism of insect resistance is crucial for insect control. As the first barrier to insecticides, insect cuticle plays an important role in insecticide resistance.. Previously, based on insectcide-resistant proteome and transcriptome research,14 cuticle protein genes were differentially expressed in deltamethrin-resistant Culex pipiens pallens campared to susceptible strain. Further study confirmed that cuticle protein gene CPLCG5 is a major gene in cuticular resistance by RNAi. In this project, by using genetic modified mosquitoes and RNAi techniques, we will further study: 1) Over-expression of CPLCG5 gene may alter the cuticle thicknesses and structures which lead to insecticide resistance; 2) The expression level of CPLCG5 gene is due to the regulation of the transcription factor Kruppel. 3) The function of CPLCG5 gene in insecticide resistance is non-specific to other types of insecticides. . The results of this project may provide insight into the molecular mechanisms of insecticide resistance and give new clues of strategy of vector control.
杀虫剂的连续、大量使用导致了虫媒抗药性的发生和发展,进而加剧了虫媒病传播的恶化趋势。研究虫媒杀虫剂抗性分子机制对虫媒防制具有重要意义。作为昆虫抵抗杀虫剂的第一道屏障,昆虫表皮对抗药性起着重要的作用。. 申请人等在前期蚊蛋白质组和转录组研究的基础上,发现14个表皮蛋白基因在淡色库蚊抗溴氰菊酯抗性品系中差异表达,采用基因微注射方法证实, CPLCG5是抗性主效表皮蛋白。本项目拟采用转基因蚊、RNAi等技术进一步研究抗性主效表皮蛋白CPLCG5可能受转录因子kruppel的调控,通过改变表皮厚度、结构发挥非特异性抗杀虫剂作用,从而揭示表皮蛋白参与杀虫剂抗性的机制及其独特的调节机制。. 本项目有望拓宽人们对媒介抗药性尤其是表皮抗性的认识,可望为媒介抗药性治理提供新靶标,并为提升现有杀虫剂的毒杀效果或开发新型杀虫剂提供新思路。
