A new type of Electrochemiluminal (ECL) cell was designed for the investigations of the ECL behavior of several reagents including N-(4-aminobutyl)-N-ethylisoluminol (ABEI), Acridinium, Daunorubicin (DNR) and Ferrocenecarboxylic acid (FCA) which were selected as markers for labeling DNA. The ECL systems for the detection of these substances were established and the ECL reaction mechanisms were also proposed on the basis of the information from the ECL, electrochemistry, fluorescence and UV measurements. Among these reagents ABEI and FCA were selected to label the known sequence oligonucleotide (DNA probe), which were then used to identify the target ssDNA immobilized on the PPY modified electrode based on the hybridization reactions between the probe and its complementary sequence. The hybridization events were evaluated by the ECL measurements. The results showed that this method allowed three-base mismatch DNA sequence to be identified and 10-11mol/L of the complementary sequence to be detected. The research of this subject offers a new method for the detection of DNA hybridization and could be used to identify of DNA sequence and determined of DNA content with high selectivity and senstivity.
本课题利用核酸分子杂化技术,研究电致化学发光活性的嵌入剂和标识剂,制备DNA电致化Х⒐馓秸耄⒏哐≡裥裕吡槊舳鹊腄NA电致化学发光分析体系,并对电致化学发光的过程及其机理进行探讨,为临床基因容要的诊断提供一种快速,简便,选择性强,灵敏度高的DNA检测新方法,并为基因芯片新的检测技术提供基础性研究。..
